【摘要】：目的:探討在匹羅卡品致癇的癲癇持續(xù)狀態(tài)(Status epilepticus,SE)大鼠模型中,Bcl-2-相互作用細(xì)胞死亡介導(dǎo)因子(Bim)與Caspase-3在大鼠海馬組織中的表達(dá)。方法:雄性成年Sprague-Dawley(SD)大鼠,應(yīng)用匹羅卡品產(chǎn)生SE 60 min后終止發(fā)作,24 h后取材,行HE染色及TUNEL染色,觀察海馬神經(jīng)元的損傷及凋亡情況,Western Blot測(cè)定海馬組織中Bim的表達(dá),免疫組化檢測(cè)Caspase-3的表達(dá)。采用Pearson相關(guān)分析法分析Bim與Caspase-3表達(dá)的相關(guān)性。結(jié)果:SE后24 h,海馬組織HE染色見(jiàn)神經(jīng)元數(shù)量減少,TUNEL染色陽(yáng)性細(xì)胞數(shù)明顯增加,Western Blot中Bim出現(xiàn)相對(duì)分子量26kD的條帶,免疫組化染色可見(jiàn)大量Caspase-3陽(yáng)性神經(jīng)元。結(jié)論:大鼠SE后24 h,Bim與Caspase-3在海馬組織神經(jīng)元表達(dá)增加,且二者的表達(dá)呈正相關(guān)(r=0.901,P=0.001),海馬神經(jīng)元出現(xiàn)凋亡。
[Abstract]:Objective: to investigate the expression of Bcl-2- and Caspase-3 in hippocampal tissue of rats with epileptic status epilepticus induced by pilocarpine. Methods: adult Sprague-Dawley SD rats were used as control group. Using pilocarpine to produce SE 60 min and stop attack 24 h later, samples were collected, HE staining and TUNEL staining were used to observe the injury and apoptosis of hippocampal neurons. Western Blot was used to detect the expression of Bim in hippocampal tissue. Immunohistochemistry was used to detect the expression of Caspase-3. Pearson correlation analysis was used to analyze the correlation between Bim and Caspase-3 expression. Results the number of neurons decreased and the number of Tunel positive cells increased significantly in hippocampal tissue 24 h after 0: SE. Bim appeared in Western Blot. Relative molecular weight of 26kD band, Conclusion: the expression of Bim and Caspase-3 in hippocampal neurons increased 24 h after SE, and there was a positive correlation between the expression of BIM and Caspase-3 in hippocampal neurons and apoptosis of hippocampal neurons.
