卒中后應(yīng)激對大鼠認(rèn)知功能與海馬MAP2表達(dá)水平的影響

【摘要】:目的:①研究卒中后應(yīng)激對大鼠認(rèn)知功能的影響。②明確卒中后應(yīng)激對大鼠海馬微管相關(guān)蛋白2(microtubule associated protein2,MAP2)表達(dá)水平的影響。 方法:①采用大腦中動(dòng)脈栓塞(middle cerebral artery occlusion,MCAO)術(shù)制備卒中模型。②抑郁癥處理采用卒中后給予慢性不可預(yù)期性溫和應(yīng)激(chronic unpredictable mild stress,CUMS)連續(xù)處理3周。③Y迷宮(Y-maze)、新穎物體識別(novel objective recognition,NOR)實(shí)驗(yàn)評估卒中后抑郁癥大鼠的認(rèn)知功能。④蛋白免疫印跡(western blot,WB)觀察海馬MAP2的表達(dá)。 結(jié)果:①與sham組(即假手術(shù)組)相比較,缺血或者慢性應(yīng)激可使大鼠體重、蔗糖偏好量、曠場實(shí)驗(yàn)水平運(yùn)動(dòng)及曠場實(shí)驗(yàn)垂直運(yùn)動(dòng)明顯下降,二者的綜合作用更明顯。②缺血或者慢性應(yīng)激可使大鼠認(rèn)知功能損傷,表現(xiàn)為與同時(shí)間點(diǎn)的sham組(即假手術(shù)組)相比較,大鼠Y迷宮試驗(yàn)的正確交替率下降和新穎物體識別試驗(yàn)中辨別指數(shù)亦下降,二者的綜合作用更明顯。③缺血或者慢性應(yīng)激可使大鼠海馬MAP2的表達(dá)下調(diào),表現(xiàn)為與sham組(即假手術(shù)組)相比較,MCAO組(即單純卒中組)和stress組(即單純抑郁癥組)大鼠海馬MAP2的表達(dá)下調(diào),二者的綜合作用PSD組(即卒中后抑郁癥組)大鼠海馬MAP2的表達(dá)下調(diào)更明顯。 結(jié)論:①卒中后應(yīng)激加重卒中大鼠認(rèn)知功能損傷;②卒中后應(yīng)激下調(diào)大鼠海馬區(qū)MAP2的表達(dá)水平。
[Abstract]:Objective to investigate the effects of post-stroke stress on cognitive function in rats. 2 to investigate the effect of post-stroke stress on the expression of microtubule-associated protein 2 associated protein 2 (MAP2) in rat hippocampus. Methods the middle cerebral artery occlusion of middle cerebral artery (MCAO) was used to make stroke model .2 depression was treated with chronic unpredictable mild stress (chronic unpredictable mild stress CUMS2) for 3 weeks. 3 Y maze to recognize novel objective. The cognitive function of poststroke depression rats was evaluated by recognition nor. The expression of hippocampal MAP2 was observed by Western blotWBs. Results compared with sham group (sham operation group), ischemia or chronic stress significantly decreased body weight, sucrose preference, horizontal exercise in open field experiment and vertical movement in open field experiment. The combined effects of the two groups were more obvious. 2. 2 Ischemia or chronic stress could induce cognitive impairment in rats, which was compared with the sham group at the same time point (that is, the sham operation group). The correct alternation rate of Y maze test decreased and the discriminant index in novel object recognition test decreased. The combined effect of these two factors was more obvious. 3. 3 ischemia or chronic stress could down-regulate the expression of MAP2 in hippocampus of rats. Compared with sham group (sham operation group), the expression of MAP2 in hippocampus was down-regulated in MCAO group (simple stroke group) and stress group (simple depression group). The down-regulation of MAP2 expression in hippocampus of PSD group (post-stroke depression group) was more obvious. Conclusion stress after stroke exacerbates cognitive impairment in stroke rats. (2) Post-stroke stress down-regulates the expression of MAP2 in hippocampal area of rats.
