【摘要】：目的探討無腦回畸形患兒的臨床特征及其致病基因。方法回顧分析2例無腦回畸形患兒的臨床表現(xiàn)、實(shí)驗(yàn)室檢查及基因檢測結(jié)果,并復(fù)習(xí)相關(guān)文獻(xiàn)。結(jié)果 2例無腦回畸形患兒均為男性,分別為7個(gè)月、3歲4個(gè)月。均以癲癇發(fā)作入院,發(fā)作時(shí)意識喪失。實(shí)驗(yàn)室檢查無明顯異常;腦電圖均提示有癲癇波;頭部MRI均提示無腦回畸形�；蚍治霭l(fā)現(xiàn),1例患兒的PAFAH1B1基因存在IVS3-1GA雜合突變,功能分析證實(shí)該位點(diǎn)突變導(dǎo)致m RNA水平外顯子4缺失,患兒父母均未見突變;另1例患兒PAFAH1B1基因存在c.274 AG突變(p.K 92 E),其父母未見異常,該突變未見報(bào)道。結(jié)論無腦回畸形患兒可能合并癲癇發(fā)生,PAFAH1B1基因突變是導(dǎo)致無腦回畸形的常見原因。
[Abstract]:Objective to investigate the clinical features and pathogenic genes in children with anencephalic gyrus malformation. Methods the clinical manifestations, laboratory and gene findings of 2 patients with anencephalic gyrus malformation were retrospectively analyzed. Results the two children with anencephalic gyrus deformity were all male, aged 3 years and 4 months, respectively. All of them were admitted to hospital with epileptic seizure, and their consciousness was lost during seizure, there was no obvious abnormality in laboratory examination, and EEG showed epileptic wave. MRI of head all showed anencephalic malformation. Gene analysis showed that there was IVS3-1GA heterozygosity in PAFAH1B1 gene of one case. Functional analysis confirmed that this mutation resulted in deletion of m RNA level exon 4, and no mutation was found in parents of the child. In the other case, there was a mutation of c.274 AG gene in the PAFAH1B1 gene, and there was no abnormal mutation in parents. Conclusion the mutation of PAFAH1B1 gene may be a common cause of anencephalic malformation in children with anencephalic malformation.
