超聲聯(lián)合微泡增效骨髓間充質(zhì)干細胞歸巢治療缺血性腦卒中

【摘要】:背景:國內(nèi)外動物實驗結(jié)果表明,超聲微泡能夠促進移植干細胞歸巢到缺血區(qū),增強血管新生和小動脈生成,改善缺血心肌局部血流量,恢復心肌收縮功能。目的:探討超聲聯(lián)合微泡對靜脈移植骨髓間充質(zhì)干細胞歸巢的影響及治療缺血性腦卒中的有效性。方法:插線法建立大腦中動脈栓塞大鼠模型,建模72 h后,隨機分為4組:PBS組(n=15),骨髓間充質(zhì)干細胞組(n=18),超聲+骨髓間充質(zhì)干細胞組(n=18),超聲+微泡+骨髓間充質(zhì)干細胞組(n=18)。PBS組經(jīng)尾靜脈注射2 mL PBS;骨髓間充質(zhì)干細胞組將骨髓間充質(zhì)干細胞(約3×10~6)用2 mL PBS稀釋后經(jīng)尾靜脈緩慢注射;超聲+骨髓間充質(zhì)干細胞組經(jīng)顱骨超聲(頻率1 MHz,功率2 W/cm~2)輻照120 s,給予骨髓間充質(zhì)干細胞;超聲+微泡+骨髓間充質(zhì)干細胞組在超聲輻照前靜注微泡對比劑0.1 mL/kg,再給予骨髓間充質(zhì)干細胞。結(jié)果與結(jié)論:(1)移植后48 h,超聲+微泡+骨髓間充質(zhì)干細胞組細胞的腦內(nèi)歸巢率高于骨髓間充質(zhì)干細胞組和超聲+骨髓間充質(zhì)干細胞組,差異均有顯著性意義(P0.05);(2)造模后28 d,超聲+微泡+骨髓間充質(zhì)干細胞組的神經(jīng)損害程度較其余組輕,差異均有顯著性意義(P0.05);(3)移植后7 d,超聲+微泡+骨髓間充質(zhì)干細胞組的腦組織含水率較其余組低,差異均有顯著性意義(P0.05);(4)移植后7 d,超聲+微泡+骨髓間充質(zhì)干細胞組腦梗死體積較其余組小,差異有顯著性意義(P0.05);(5)結(jié)果表明,超聲聯(lián)合微泡可促進骨髓間充質(zhì)干細胞的腦內(nèi)靶向歸巢,能夠減少腦水腫和腦梗死體積,改善神經(jīng)功能評分,增效骨髓間充質(zhì)干細胞移植治療缺血性腦卒中的效果。
[Abstract]:Background: the results of animal experiments at home and abroad show that ultrasound microbubbles can promote the homing of stem cells to ischemic areas, enhance angiogenesis and arteriogenesis, and improve the local blood flow of ischemic myocardium. Objective: to investigate the effect of ultrasound combined with microbubbles on homing of bone marrow mesenchymal stem cells (BMSCs) after vein transplantation and the efficacy in the treatment of ischemic stroke. 72 hours after modeling, They were randomly divided into 4 groups: PBS group (n = 15), bone marrow mesenchymal stem cell group (n = 18), ultrasound bone marrow mesenchymal stem cell group (n ~ (18)), ultrasound microbubble bone marrow mesenchymal stem cell group (n ~ (18)) and PBS group (n = 2 mL) were injected with 2 mL PBSvia tail vein; bone marrow mesenchymal stem cell group (BMSCs) was injected with bone marrow. Mesenchymal stem cells (3 脳 10 6) were diluted with 2 mL PBS and injected slowly through the tail vein. Bone marrow mesenchymal stem cells (BMSCs) were irradiated with ultrasound for 120 s (frequency 1 MHz, power 2 W / cm 2) for 120 s, and bone marrow mesenchymal stem cells (BMSCs) were given. Ultrasound microbubble bone marrow mesenchymal stem cells group was given bone marrow mesenchymal stem cells by intravenous injection of 0.1 mL / kg contrast agent before ultrasound irradiation. Results and conclusion 48 hours after transplantation, ultrasound microbubble bone marrow mesenchymal stem cells were treated with ultrasound microbubble bone marrow mesenchymal stem cells. The homing rate in brain was higher than that in bone marrow mesenchymal stem cells group and ultrasound bone marrow mesenchymal stem cell group. There was significant difference between the two groups at 28 days after the model was made, the nerve damage in the ultrasound microbubble bone marrow mesenchymal stem cells group was lighter than that in the other groups. At 7 days after transplantation, the water content of brain tissue in the ultrasound microbubble bone marrow mesenchymal stem cells group was lower than that in the other groups. At 7 days after transplantation, the volume of cerebral infarction in the ultrasound microbubble bone marrow mesenchymal stem cells group was smaller than that in the other groups, and the difference was significant (P 0.05). Ultrasound combined with microbubbles can promote the homing of bone marrow mesenchymal stem cells, reduce the volume of brain edema and cerebral infarction, improve the neurological function score, and enhance the effect of bone marrow mesenchymal stem cell transplantation in the treatment of ischemic stroke.
