比較額顳葉及小腦皮層電驚厥閾值與神經(jīng)損害的實(shí)驗(yàn)研究

【摘要】:目的:通過建立大鼠的皮層驚厥閾值模型測定并比較額葉、顳葉及小腦皮層的驚厥閾值;比較額顳葉及小腦皮層驚厥閾值穩(wěn)定后的海馬CA3區(qū)神經(jīng)元變化。方法:選擇健康雌性SD大鼠,體重180-220g之間,分別在額葉皮層、顳葉皮層、小腦皮層安放電極造模,作為刺激組,對照組僅安放電極不予刺激,使用直流電每日2次刺激大鼠額葉、顳葉、小腦皮層,當(dāng)刺激組驚厥閾值穩(wěn)定后,處死各組大鼠,用Nissl染色法觀察海馬神經(jīng)元損害情況。結(jié)果:1、實(shí)驗(yàn)開始時(shí),大鼠三個(gè)刺激組皮層的驚厥閾值均很高,額葉組閾值波動在1100μA上下,顳葉組閾值波動在1200μA上下,小腦組閾值波動在2100μA上下,隨著刺激時(shí)間的延長,各組大鼠的皮層閾值不斷降低,并最終穩(wěn)定在一個(gè)基線水平,但其基線各不相同,額葉組閾值波動于520μA左右,顳葉組穩(wěn)定在440μA左右,小腦組穩(wěn)定在1300μA左右,通過單因素方差分析,比較各組穩(wěn)定后的驚厥閾值,額顳葉組的P0.05,小腦與額顳葉組的P0.05;2、隨著刺激時(shí)間的延長,刺激組大鼠腦海馬CA3區(qū)神經(jīng)元呈現(xiàn)不同程度的缺失,且排列紊亂,對照組高倍鏡下觀察神經(jīng)元數(shù)目約(36.33±6.13)個(gè),額葉組、顳葉組、小腦組神經(jīng)元數(shù)目分別為:(22.79±1.01)個(gè)、(17.78±1.40)個(gè)、(26.00±1.70)個(gè),與對照組比較,額葉、顳葉、小腦組神經(jīng)元個(gè)數(shù)減少,三組間比較具有統(tǒng)計(jì)學(xué)差異(P0.05);3、電刺激小腦皮層時(shí)大鼠出現(xiàn)的癥狀與額、顳葉皮層的表現(xiàn)不盡相同,額顳葉組均表現(xiàn)出典型的癇樣抽搐,如點(diǎn)頭、動須、四肢及全身強(qiáng)直-陣攣行為,而小腦組表現(xiàn)為全身的強(qiáng)直痙攣以及奔跑、跳躍等。結(jié)論:1、顳葉與額葉皮層對電刺激比小腦皮層更敏感,更容易形成癲癇病灶,而顳葉與額葉之間對刺激的易感性大致相同;2、海馬神經(jīng)元損害嚴(yán)重程度:顳葉額葉小腦。
[Abstract]:Objective: to determine and compare the threshold of convulsion in frontal lobe temporal lobe and cerebellar cortex by establishing a rat model of cortical convulsion threshold. To compare the changes of hippocampal CA3 neurons in frontal temporal lobe and cerebellar cortex after the convulsion threshold was stabilized. Methods: healthy female SD rats weighing 180-220 g were selected respectively in frontal cortex and temporal cortex. The cerebellar cortex was set up as a model, and the control group was treated only with electrodes without stimulation. The frontal lobe, temporal lobe and cerebellar cortex were stimulated by DC twice a day, when the threshold of convulsion was stable in the stimulation group. The damage of hippocampal neurons was observed by Nissl staining. Results: at the beginning of the experiment, the cortical convulsion thresholds of the three stimulation groups were all very high. The threshold of frontal lobe group fluctuated around 1100 渭 A, temporal lobe group about 1200 渭 A, cerebellar group about 2100 渭 A, with the extension of stimulation time. The cortical threshold of each group decreased continuously, and finally stabilized at a baseline level, but the baseline was different. The threshold of frontal lobe group fluctuated around 520 渭 A, and that of temporal lobe group was about 440 渭 A. The cerebellar group was stable at about 1300 渭 A. the threshold of convulsion was compared between the frontotemporal lobe group and the cerebellar and frontotemporal lobe group by univariate ANOVA. 2, with the prolongation of the stimulation time, the neurons in the CA3 area of the brain of the rats in the stimulation group showed different degrees of deletion and disordered arrangement. The number of neurons was about 36.33 鹵6.13 in the control group, 22.79 鹵1.01 in the frontal lobe group, temporal lobe group and cerebellar group, respectively. Compared with the control group, the number of neurons in the frontal lobe, temporal lobe and cerebellum decreased. There was statistical difference among the three groups (P 0.05). 3. The symptoms of cerebellar cortex were different from those of frontotemporal cortex. In frontotemporal lobe group, typical epileptiform convulsions were observed, such as nodding, motor whiskers, limbs and whole body tonic-clonic behavior. The cerebellar group showed systemic ankylosis spasm, running and jumping. Conclusion 1, temporal lobe and frontal cortex are more sensitive to electrical stimulation than cerebellar cortex, and epileptic foci are more easily formed. The susceptibility to stimulation was similar between temporal lobe and frontal lobe. 2, the severity of hippocampal neuron damage: temporal frontal lobe cerebellum.
