人腦膠質(zhì)瘤中ALDH1表達(dá)及其與Ki-67和MGMT的關(guān)系研究

【摘要】:目的研究ALDH1、Ki-67和MGMT在人腦膠質(zhì)瘤組織中的表達(dá)與膠質(zhì)瘤病人的臨床資料的關(guān)系;探討ALDH1、Ki-67和MGMT三者之間的關(guān)系,并闡明它們與膠質(zhì)瘤惡性程度的關(guān)系;聯(lián)合檢測(cè)ALDH1、Ki-67和MGMT蛋白,為準(zhǔn)確地診斷膠質(zhì)瘤的惡性程度提供一種方法,同時(shí),也為膠質(zhì)瘤的個(gè)體化治療提供理論依據(jù)。 方法連續(xù)收集蘭州大學(xué)第二附屬醫(yī)院神經(jīng)外科臨床中心2012年7月至2013年10月間經(jīng)手術(shù)治療的膠質(zhì)瘤病人的病理標(biāo)本及臨床資料53例,納入研究,所有病例均為新發(fā)診斷為膠質(zhì)瘤的病人,排除復(fù)發(fā)的病例、以及之前施行化療、放療等治療的病例。采用免疫組織化學(xué)的方法檢測(cè)膠質(zhì)瘤標(biāo)本中ALDH1、Ki-67和MGMT蛋白的表達(dá)情況,采用SPSS18.0統(tǒng)計(jì)學(xué)軟件分析ALDH1、Ki-67和MGMT蛋白在人腦膠質(zhì)瘤組織中的表達(dá)及意義。 結(jié)果本研究顯示,患者的性別、年齡及腫瘤組織學(xué)類型與膠質(zhì)瘤組織中ALDH1、Ki-67和MGMT的表達(dá)無關(guān)。高級(jí)別組(WHOⅢ-Ⅳ級(jí))中ALDH1、Ki-67和MGMT的表達(dá)均較低級(jí)別組(WHO Ⅰ-Ⅱ級(jí))顯著升高(ALDH1,93.10%比70.83%,p0.01;Ki-67,96.55%比70.83%,p0.01;MGMT,72.41%比50.00%,p0.01)。另外,本研究發(fā)現(xiàn)人腦膠質(zhì)瘤中ALDH1、Ki-67和MGMT三者之間呈高度顯著正相關(guān)關(guān)系(ALDH1與Ki-67, rs=O.904, p=0.000; ALDH1與MGMT, rs=0.833, p=0.000; Ki-67與MGMT, rs=0.779, p=0.000)。 結(jié)論結(jié)果表明,在人腦膠質(zhì)瘤中,ALDH1,Ki-67和MGMT蛋白的表達(dá)與膠質(zhì)瘤的病理級(jí)別相關(guān),級(jí)別越高,三種蛋白的表達(dá)就越強(qiáng)。ALDH1、Ki-67和MGMT三者之間高度顯著正相關(guān)關(guān)系,表明膠質(zhì)瘤中ALDH1、Ki-67和MGMT蛋白三者之間的內(nèi)在一致性聯(lián)系,聯(lián)合檢測(cè)三個(gè)指標(biāo)可能更能準(zhǔn)確地判斷膠質(zhì)瘤患者惡性程度和預(yù)后,對(duì)膠質(zhì)瘤患者制定個(gè)體化的治療方案可能有著重要的參考價(jià)值。
[Abstract]:Objective to study the relationship between the expression of ALDH1 Ki-67 and MGMT in human glioma tissues and the clinical data of glioma patients. To explore the relationship between Ki-67 and MGMT in ALDH1, and to clarify their relationship with the malignant degree of glioma. The combined detection of ALDH1 Ki-67 and MGMT protein provides a method for the accurate diagnosis of malignant degree of glioma and also provides theoretical basis for individualized treatment of glioma. Methods the pathological specimens and clinical data of 53 patients with glioma treated surgically from July 2012 to October 2013 in the Neurosurgery Clinical Center of the second affiliated Hospital of Lanzhou University were collected. Included in the study, all cases were newly diagnosed as gliomas, excluding recurrent cases, and prior chemotherapy. Immunohistochemical method was used to detect the expression of ALDH1Ki-67 and MGMT protein in gliomas. The expression and significance of ALDH1 Ki-67 and MGMT protein in human gliomas were analyzed by SPSS18.0 software. Results in this study, sex, age, tumor histological type and ALDH1 in glioma tissues were found.
