丘腦底核深部腦刺激改善帕金森病模型大鼠自發(fā)跑輪行為的研究

【摘要】:目的探討丘腦底核(subthalamic nucleus,STN)深部腦刺激(deep brain stimulation,DBS)能否改善6-羥基多巴胺偏側(cè)帕金森病模型大鼠自發(fā)跑輪行為。方法將35只Sprague-Dawley(SD)大鼠隨機分為實驗組24只和對照組11只。利用立體定向的方法將刺激電極埋入右側(cè)丘腦底核。實驗組大鼠右側(cè)內(nèi)側(cè)前腦束內(nèi)注射12μg 6-OHDA,對照組注射相同體積的生理鹽水。對造模前、造模后和給予電刺激時的大鼠進行前肢使用不對稱測試和自發(fā)跑輪行為測試。酪氨酸羥化酶免疫組化染色檢測黑質(zhì)多巴胺能神經(jīng)元毀損程度。蘇木素-伊紅染色確定刺激電極尖端位置。結(jié)果從24只大鼠中獲得了17只成功的PD大鼠模型。前肢使用不對稱測試:與毀損后未給予STN-DBS時比較,電極尖端位于STN內(nèi)的成功模型大鼠給予STN-DBS后,不對稱指數(shù)顯著下降(P=0.038),而與毀損前相比,差異不顯著(P=0.07)。跑輪行為測試:與毀損后未給予STN-DBS時比較,電極尖端位于STN內(nèi)的成功模型大鼠給予STN-DBS后,跑動次數(shù)(P=0.032)、跑動距離(P=0.03)和平均峰速度均(P=0.035)顯著增加,而從跑動開始到峰速度的平均時間顯著減小(P=0.001)。這些指標與毀損前比較,差異均無統(tǒng)計學(xué)意義。結(jié)論丘腦底核深部腦刺激能夠改善6-羥基多巴胺偏側(cè)帕金森病模型大鼠的自發(fā)跑輪行為,自發(fā)跑輪行為測試在一定程度上可用于篩選抗帕金森病藥物或其他治療方法。
[Abstract]:Objective to investigate the deep brain stimulation of the subthalamic nucleus subthalamic nucleus (STN). Methods 35 SD rats with Sprague-Dawley disease were treated with DBS to improve the spontaneous wheel running behavior of 6-hydroxydopamine hemiparkinsonism model rats. Rats were randomly divided into experimental group (n = 24) and control group (n = 11). The stimulation electrode was implanted into the right subthalamic nucleus by stereotactic method. In the experimental group, 12 渭 g 6-OHDA was injected into the right medial forebrain tract. The control group was injected with the same volume of normal saline. The antecedent limb was tested by asymmetrical test and spontaneous wheel running behavior test after modeling and electrical stimulation. Tyrosine hydroxylase immunohistochemical staining was used to detect the damage degree of substantia nigra dopaminergic neurons. Hematoxylin-eosin staining. Results 17 successful PD rat models were obtained from 24 rats. The results were compared with those when STN-DBS was not given after damage. After STN-DBS was given to the successful model rats with the tip of the electrode located in STN, the asymmetry index decreased significantly (P < 0.05), compared with that before STN-DBS. Test of running wheel behavior: compared with those without STN-DBS, the successful model rats with tip of electrode located in STN were given STN-DBS. The running times (P0. 032), the running distance (P0. 03) and the average peak velocity (P0. 035) increased significantly. However, the average time from the beginning of running to the peak velocity decreased significantly. These indexes were compared with those before damage. Conclusion the deep brain stimulation of subthalamic nucleus can improve the spontaneous wheel running behavior of 6-hydroxydopamine hemiparkinsonism model rats. Spontaneous wheel-running behavior testing can be used to screen antiParkinson disease drugs or other treatment methods to some extent.
