脊神經(jīng)結(jié)扎大鼠脊髓背角結(jié)構(gòu)可塑性的體視學(xué)研究

【摘要】:目的:探討L5脊神經(jīng)緊結(jié)扎(SNL)所致神經(jīng)病理性疼痛(NP)對大鼠脊髓背角內(nèi)神經(jīng)元、突觸、小膠質(zhì)細(xì)胞及星形膠質(zhì)細(xì)胞數(shù)量的影響。方法:將29只3月齡雄性SD(Sprague-Dawley)大鼠隨機(jī)分為2組:SNL組(n=16,行單側(cè)L5脊神經(jīng)緊結(jié)扎)和假手術(shù)組(n=13,行單側(cè)假手術(shù))。每組分別于術(shù)后7天和28天切取L5節(jié)段脊髓進(jìn)行石蠟包埋并制作14μm厚的連續(xù)切片50張,采用等距隨機(jī)抽樣原則分別各抽選3-5張用尼氏染色以及Synaptophysin、IBa-1、GFAP抗體的免疫組織化學(xué)染色顯示神經(jīng)元、突觸素顆粒、小膠質(zhì)細(xì)胞及星形膠質(zhì)細(xì)胞。采用體視學(xué)新技術(shù)——光學(xué)體視框估計(jì)脊髓背角內(nèi)神經(jīng)元、突觸、小膠質(zhì)細(xì)胞、星形膠質(zhì)細(xì)胞的數(shù)量的變化結(jié)果:與對側(cè)未手術(shù)側(cè)相比,SNL組手術(shù)側(cè)術(shù)后7 d單位長度L5節(jié)段脊髓背角內(nèi)的突觸數(shù)及突觸數(shù)與神經(jīng)元數(shù)之比均無顯著變化,小膠質(zhì)細(xì)胞及星形膠質(zhì)細(xì)胞數(shù)量顯著增加了210%和62%;術(shù)后28 d,突觸數(shù)及突觸數(shù)與神經(jīng)元數(shù)之比分別顯著增加了29%和20%,小膠質(zhì)細(xì)胞的數(shù)量顯著增加了120%,星形膠質(zhì)細(xì)胞數(shù)量無顯著變化。與對側(cè)未手術(shù)側(cè)相比,假手術(shù)組手術(shù)側(cè)神經(jīng)元、突觸、小膠質(zhì)細(xì)胞和星形膠質(zhì)細(xì)胞的數(shù)量術(shù)后7d和28 d均無顯著性變化。SNL組手術(shù)側(cè)術(shù)后28 d脊髓背角內(nèi)的小膠質(zhì)細(xì)胞數(shù)與星形膠質(zhì)細(xì)胞數(shù)與7 d相比分別顯著減少了48%和20%。結(jié)論:L5脊神經(jīng)結(jié)扎導(dǎo)致大鼠脊髓背角發(fā)生結(jié)構(gòu)上的可塑性變化,該變化隨著NP的進(jìn)展呈現(xiàn)出時(shí)間相關(guān)性的動態(tài)趨勢:突觸數(shù)量呈現(xiàn)由無顯著增加到逐漸出現(xiàn)顯著增加的變化趨勢;膠質(zhì)細(xì)胞數(shù)量則呈現(xiàn)早期極顯著增加,隨后逐漸減少的變化趨勢。小膠質(zhì)細(xì)胞和星形膠質(zhì)細(xì)胞可能與外周神經(jīng)損傷早期NP的誘導(dǎo)和產(chǎn)生有關(guān),突觸可能與NP的維持有關(guān)。
[Abstract]:Objective: To investigate the L5 spinal nerve tight ligation (SNL) induced neuropathic pain (NP) synapses on rat spinal cord dorsal horn neurons, and affect the number of microglia and astrocytes. Methods: 29 March old male SD (Sprague-Dawley) rats were randomly divided into 2 groups: SNL group (n=16. Unilateral tight ligation of L5 spinal nerve) and sham operation group (n=13, unilateral sham). Each group in 7 days and 28 days after the L5 segment of the spinal cord were embedded in paraffin and made 14 consecutive sections with the thickness of M 50, using a uniform random sampling principle, all selected by 3-5 Zhang Nissl staining and Synaptophysin, IBa-1, immunohistochemistry of GFAP antibody staining showed that synaptophysin granules, neurons, astrocytes and microglia. Using stereological technique -- optical disector estimation in the spinal dorsal horn neurons, synapses, microglial cells, the number of astrocytes The change results: compared with the contralateral side of surgery, the operative side in group SNL after 7 d per unit length of L5 segment of the spinal cord in the dorsal horn of the number and the number of synapses and synaptic neuron number ratio did not change significantly, the number of microglia and astrocytes was significantly increased by 210% and 62%; 28 d after operation the number, the number of synapses and synaptic and neuronal number ratio were increased by 29% and 20%, the number of microglia cells were significantly increased by 120%, the number of astrocytes. No significant changes compared with the contralateral unoperated side, sham operated group, neurons, synapses, the number of microglia and astrocytes. After 7d and 28 d were no significant changes in.SNL operated group after 28 d in the spinal dorsal horn microglia and astrocytes number compared with 7 d were significantly reduced by 48% and 20%. conclusion: L5 spinal nerve ligation in the rat spinal dorsal horn structure can occur The plastic changes, the changes with the progress of NP shows a dynamic trend time correlation: the number of synapses presented by no significant increase trend to gradually increased significantly; number of glial cells showed a significant increase trend in early stage, and then decreased gradually. Microglia and astrocytes and peripheral nerve the early damage induced by NP and related synapses may be related to NP.
