【摘要】：目的：通過研究酸脂清不同提取物對痛風(fēng)性關(guān)節(jié)炎模型大鼠血清IL-1β、TNF-α、IL-10、IL-1Ra濃度、關(guān)節(jié)腫脹和關(guān)節(jié)滑膜病理變化的影響，，評價酸脂清對痛風(fēng)性關(guān)節(jié)炎的抗炎和關(guān)節(jié)保護(hù)作用。 方法：將80只實(shí)驗(yàn)用Wistar大鼠（200±20g,雄性），隨機(jī)分成空白組、模型組、水提酸脂清組、醇提酸脂清組和秋水仙堿組5組，每組16只，空白組和模型組給予生理鹽水灌胃，水提酸脂清組、醇提酸脂清組和秋水仙堿組分別給予酸脂清水提物、醇提物和秋水仙堿片混懸液灌胃,第7天灌胃結(jié)束后0.5h，測量大鼠右踝關(guān)節(jié)直徑，并選定踝關(guān)節(jié)外側(cè)后方以尿酸鈉晶體混懸液局部注射造成痛風(fēng)性關(guān)節(jié)炎模型。造模后9h、12h在各組中隨機(jī)抽取8只，測量造模踝關(guān)節(jié)直徑；麻醉后下腔靜脈取血，Elisa法檢測IL-1β、TNF-α、IL-10、IL-1Ra濃度；將造模踝關(guān)節(jié)取下，做病理切片觀察滑膜炎癥變化。 結(jié)果：①兩酸脂清水提取物組較模型組腫脹程度明顯降低，有顯著性意義（P＜0.05）；②三個藥物組與模型組比較，IL-1β、TNF-α（軟骨破壞因子）濃度明顯降低、IL-10、IL-1Ra（保護(hù)因子）濃度明顯升高（P＜0.05）；③三個藥物組關(guān)節(jié)滑膜炎癥與模型組關(guān)節(jié)滑膜炎癥比較，三個藥物組滑膜炎癥明顯輕于模型組；④酸脂清水提組可明顯升高IL-10濃度，與其他各組比較有顯著性差異（P＜0.05）。 結(jié)論：酸脂清通過減輕痛風(fēng)性關(guān)節(jié)炎模型大鼠關(guān)節(jié)炎癥，降低關(guān)節(jié)軟骨破壞因子IL-1β和TNF-α濃度，升高軟骨保護(hù)因子IL-10和IL-1Ra的濃度，達(dá)到治療痛風(fēng)性關(guān)節(jié)炎，減少關(guān)節(jié)損傷的目的。其中酸脂清水提物對IL-10的升高效果較明顯，故酸脂清水提物效果較好。
[Abstract]:Aim: to study the effects of different extracts of acid Zhiqing on serum IL-1 尾, TNF- 偽, IL-10,IL-1Ra concentration, joint swelling and pathological changes of synovium in rats with gouty arthritis, and to evaluate the anti-inflammatory and joint protective effects of acid Zhiqing on gouty arthritis. Methods: 80 Wistar rats (200g 鹵20g, male) were randomly divided into three groups: blank group, model group, water extract group, alcohol extract group and colchicine group, with 16 rats in each group. The blank group and model group were given saline intragastrically, water extract group, alcohol extract group and colchicine group were given acid lipid extract, alcohol extract and colchicine tablet suspension respectively. 0.5 h after the end of gastric administration on the 7th day, the rats in the blank group and the model group were given saline, water extract, alcohol extract and colchicine tablet suspension, respectively, and the rats in the blank group and the model group were given saline, water extract, alcohol extract and colchicine tablet suspension respectively. The diameter of right ankle joint was measured and the model of gouty arthritis was established by local injection of sodium urate crystal suspension. At 9 h and 12 h after modeling, 8 rats were randomly selected from each group to measure the diameter of malleolus joint, the blood was taken from inferior vena cava after anesthesia, the concentrations of IL-1 尾, TNF- 偽 and IL-10,IL-1Ra were detected by Elisa method, and the malleolus joint was removed and pathological section was made to observe the changes of synovitis. Results: (1) the swelling degree of the extract group was significantly lower than that of the model group (P < 0.05), and the concentration of IL-1 尾, TNF- 偽 (cartilage destruction factor) and IL-10,IL-1Ra (protective factor) in the three drug groups were significantly lower than those in the model group (P < 0.05). 3Compared with the model group, the synovitis in the three drug groups was significantly lighter than that in the model group, and the concentration of IL-10 in the acid extract group was significantly higher than that in the other groups (P < 0.05). Conclusion: Zaozhiqing can treat gouty arthritis and reduce joint injury by reducing arthritis, decreasing the concentration of IL-1 尾 and TNF- 偽, and increasing the concentration of cartilage protective factor IL-10 and IL-1Ra. The effect of acid-fat clear water extract on the increase of IL-10 was obvious, so the effect of acid-lipid clean water extract was better.
