【摘要】：目的:探討NCAM在大鼠腦缺血再灌注后不同時(shí)間的表達(dá)變化及芹菜素對(duì)其表達(dá)的影響。方法:采用改良線栓法建立大鼠腦缺血再灌注后不同時(shí)間損傷模型。動(dòng)物隨機(jī)分為假手術(shù)組(S)、模型組(M)、芹菜素組(A)和地塞米松組(D)。各手術(shù)組按再灌注時(shí)間不同又分為再灌注6 h組、24 h組、72 h組、7 d組,各組動(dòng)物麻醉清醒后均給予神經(jīng)行為學(xué)評(píng)分,每組1只大鼠TTC染色觀察腦梗死灶、6只免疫組化法測(cè)定NCAM的表達(dá)。結(jié)果在A72h組神經(jīng)行為學(xué)評(píng)分低于M72h(P0.05);腦組織切片TTC染色顯示各手術(shù)組均出現(xiàn)白色梗死灶;正常腦組織NCAM少量表達(dá),缺血再灌注后表達(dá)增加,于72 h達(dá)高峰,A組能顯著提高NCAM的表達(dá),各時(shí)間點(diǎn)A和M組比較差異顯著(P0.05)。結(jié)論:芹菜素可增加腦缺血再灌注后NCAM的表達(dá),提示芹菜素可能通過(guò)NCMA達(dá)到促進(jìn)腦組織損傷后神經(jīng)細(xì)胞修復(fù)和再生的作用。
[Abstract]:Aim: to investigate the changes of NCAM expression at different time after cerebral ischemia reperfusion in rats and the effect of apigenin on it. Methods: the rat model of cerebral ischemia reperfusion injury at different time was established by modified thread embolization. Animals were randomly divided into sham operation group (S), model group (M), apigenin group (A) and dexamethasone group (D). Each operation group was divided into 6 h group, 24 h group, 72 h group and 7 day group according to the time of reperfusion. The rats in each group were given neurobehavioral score after anaesthesia. The cerebral infarction was observed by TTC staining in each group. The expression of NCAM was detected by immunohistochemical method in 6 mice. Results the neurobehavioral score in A72 h group was lower than that in M72 h group (P0.05), and the white infarct foci appeared in all the operation groups by TTC staining. The expression of NCAM in normal brain tissue increased after ischemia reperfusion and reached the peak at 72 hours. Group A could significantly increase the expression of NCAM, and the difference was significant between group A and group M at each time point (P0.05). Conclusion: apigenin can increase the expression of NCAM after cerebral ischemia-reperfusion, suggesting that apigenin can promote the repair and regeneration of nerve cells after brain injury through NCMA.
