白芍木瓜湯防治大鼠骨性關(guān)節(jié)炎的實驗研究

【摘要】:目的:建立骨性關(guān)節(jié)炎大鼠模型,用白芍木瓜湯治療干預(yù),觀察白芍木瓜湯對OA大鼠一般情況、體重、抓力、影像學和病理學表現(xiàn)的影響,驗證白芍木瓜湯對OA大鼠防治作用,并探討其對骨性關(guān)節(jié)炎的可能作用機制。方法:清潔級雄性SD大鼠,共50只,將其隨機分成5組:正常對照組(A組)、模型對照組(B組)、陽性對照組(C組)、白芍木瓜湯高劑量組(D組)、白芍木瓜湯低劑量組(E組),每組10只。其中正常對照組不予任何處理,其余4組大鼠均采用雙側(cè)膝關(guān)節(jié)腔內(nèi)注射碘乙酸法,成功建立OA大鼠模型。術(shù)后第二天,A、B組給予生理鹽水灌胃,C組給予布洛芬灌胃,D、E兩組采用不同劑量的中藥湯劑灌胃,持續(xù)灌胃6周,在此期間,每周稱量各組大鼠的體重及測定其抓力程度。至第6周末,對各組大鼠行腹腔麻醉,用X-ray影像觀察膝關(guān)節(jié)結(jié)構(gòu)與狀態(tài);心臟采血,并分離血清,用酶聯(lián)免疫(Elisa)法測定各組大鼠的血清中基質(zhì)金屬蛋白酶13(MMP-13)、軟骨蛋白聚糖(Aggrecan)、Ⅱ型膠原(Col Ⅱ)含量;并截取膝關(guān)節(jié),剔除皮膚與肌肉,EDTA脫鈣后分別進行HE染色與SO染色,光鏡下觀察各組大鼠膝關(guān)節(jié)病理變化。結(jié)果:(1)體重:造模成功后,B組的大鼠體重相對于A組,始終存在顯著性差異(P0.01),隨著給藥時間的增加,白芍木瓜湯可以改善大鼠食欲,使大鼠體重增加。(2)抓力測定:各治療組的大鼠抓力水平始終無明顯差異(P0.05)。(3)膝關(guān)節(jié)影像:造模成功后,相對于A組,B組大鼠的雙膝關(guān)節(jié)破壞嚴重,干預(yù)治療6周后,C、D、E組大鼠雙膝關(guān)節(jié)的影像學表現(xiàn)均明顯改善,以D組的改善情況最為明顯。(4)膝關(guān)節(jié)病理:造模成功后,相比于A組,B組的大鼠雙膝關(guān)節(jié)破壞程度高;干預(yù)治療6周后,相比于B組,C、D、E組膝關(guān)節(jié)軟骨的病理學均明顯改善,以D組的改善情況最為明顯。(5)血清含量:造模成功后,相對于A組,B組大鼠血清中的MMP-13含量呈顯著性升高(P0.01),Col-Ⅱ、Aggrecan含量呈顯著性降低(P0.01);藥物治療6周后,相比于B組,C、D、E組大鼠血清中的MMP-13含量均呈顯著性降低(P0.01),血清Col-Ⅱ、Aggrecan含量均呈顯著性升高(P0.01)。結(jié)論:白芍木瓜湯對碘乙酸誘導(dǎo)的大鼠骨性關(guān)節(jié)炎有良好的藥效作用,其作用機制可能與抑制MMP-13產(chǎn)生,促進Aggrecan、Col Ⅱ合成,從而達到修復(fù)軟骨有關(guān)。
[Abstract]:Objective: to establish a rat model of osteoarthritis, and to observe the effects of paeonia-papaya decoction on the general condition, body weight, grasping ability, imaging and pathology of OA rats, and to verify the preventive and therapeutic effects of paeonia-papaya decoction on OA rats. To explore the possible mechanism of its action on osteoarthritis. Methods: fifty clean grade male SD rats were used. They were randomly divided into 5 groups: normal control group (group A), model control group (group B), positive control group (group C), high dose group of paeoniae papaya decoction (group D) and low dose group of paeoniao papaya decoction (group E) with 10 rats in each group. The normal control group was not treated with any treatment, and the other four groups were successfully established OA rat model by injecting iodoacetic acid into bilateral knee joint. On the second day after operation, normal saline was given in group A and ibuprofen was given in group C. The rats in group D were given different doses of traditional Chinese medicine decoction for 6 weeks. During this period, the weight of rats in each group was measured weekly and the degree of grasping was measured. At the end of the 6th week, the rats in each group were anesthetized intraperitoneally, the structure and state of knee joint were observed by X-ray imaging, the blood was taken from the heart, and the serum was separated.
