【摘要】：目的:觀察新型腎上腺素α,β受體雙重阻斷劑TJ0711靜脈注射對(duì)自發(fā)性高血壓大鼠(SHR)和哇巴因-高血壓大鼠(OHR)2種模型的急性降壓作用。方法:制備OHR模型:SD大鼠連續(xù)8周腹腔注射30μg·kg-1·d-1哇巴因,將收縮壓21.3 k Pa(1 k Pa≈7.5 mm Hg)者隨機(jī)分為生理鹽水(N.S)組、TJ0711 0.3,0.9,2.7 mg·kg-1劑量組、陽性對(duì)照卡維地洛1 mg·kg-1組(n=12);購(gòu)得的SHR分組方法同OHR。用10%烏拉坦(1.3 g·kg-1,i.p)麻醉,仰位固定,分離一側(cè)頸總動(dòng)脈插管連接多通道生理記錄儀測(cè)血壓和心率,頸外靜脈插管給藥。觀察給藥前后大鼠收縮壓、舒張壓、心率的變化,分析TJ0711靜注降壓作用規(guī)律。結(jié)果:TJ0711靜注給藥后2~15 min即達(dá)到最大降壓水平,降壓作用維持25~75 min。對(duì)SHR和OHR兩種高血壓模型收縮壓與舒張壓下降趨勢(shì)一致,但對(duì)SHR收縮壓、舒張壓最大降壓幅度高于OHR。結(jié)論:TJ0711靜脈注射能快速、短效降低SHR和OHR兩高血壓模型大鼠的血壓同時(shí)使心率適當(dāng)減慢,具有良好的急性降壓特點(diǎn)。
[Abstract]:Objective: To observe the acute antihypertensive effect of a new type of adrenalin alpha and beta receptor double blocker TJ0711 on 2 models of spontaneously hypertensive rats (SHR) and ouabain hypertensive rats (OHR). Methods: to prepare a OHR model: SD rats were intraperitoneally injected with 30 u g. Kg-1. D-1 ouabain for 8 weeks, and the systolic pressure was 21.3 K Pa (1 K Pa 7.5). The machine was divided into two groups: normal saline (N.S) group, TJ0711 0.3,0.9,2.7 mg / kg-1 dose group, positive control carvedilol 1 mg kg-1 group (n=12). The SHR grouping method and OHR. were anaesthetized with 10% urantan (1.3 g kg-1), and the side cervical artery intubation was attached to the multichannel physiological recorder to measure blood pressure and heart rate, and the external jugular vein catheterization was given. The changes of systolic pressure, diastolic pressure and heart rate of rats before and after administration were observed. The results were as follows: 2~15 min reached the maximum depressor level after intravenous injection of TJ0711, and the antihypertensive effect maintained 25~75 min. on the decreasing trend of systolic pressure and diastolic pressure of two kinds of hypertension models of SHR and OHR, but the maximum decrease of systolic pressure and diastolic pressure on SHR systolic pressure. The pressure amplitude is higher than the OHR. conclusion: the intravenous injection of TJ0711 can quickly and effectively reduce the blood pressure of the rat model of SHR and OHR two, and the heart rate can be slowed down properly, and it has a good characteristic of acute hypotension.
