帕瑞昔布對(duì)肝部分切除術(shù)老年大鼠海馬炎癥反應(yīng)的影響

【摘要】:目的研究帕瑞昔布對(duì)肝部分切除術(shù)老年大鼠海馬炎癥反應(yīng)的影響。方法選擇SPF級(jí)雄性SD大鼠70只,20月齡,體重500~600g,隨機(jī)分為三組:對(duì)照組(C組,n=10),手術(shù)組(S組,n=30)及帕瑞昔布組(P組,n=30)。采用肝部分切除術(shù)建立手術(shù)創(chuàng)傷導(dǎo)致海馬炎癥反應(yīng)模型。P組大鼠在異氟醚麻醉下行肝部分切除術(shù),并于術(shù)前1h腹腔注射帕瑞昔布10 mg/kg,術(shù)后每日腹腔注射帕瑞昔布2mg/kg直至處死。S組大鼠在異氟醚麻醉下行肝部分切除術(shù),術(shù)前1h腹腔注射與P組等量的生理鹽水。C組腹腔注射與以上各組等量的生理鹽水。干預(yù)后第1、3、7天處死大鼠,取海馬組織,采用實(shí)時(shí)定量PCR法檢測(cè)環(huán)氧合酶-2(COX-2)mRNA的表達(dá)量,Western blot法檢測(cè)COX-2、半胱氨酸蛋白酶-3(caspase-3)的蛋白表達(dá)量,ELISA法檢測(cè)前列腺素E2(PGE2)的含量。結(jié)果干預(yù)后第1、3天S組和P組COX-2mRNA及蛋白表達(dá)、PGE2含量、caspase-3蛋白表達(dá)明顯高于C組(P0.05或P0.01),但P組明顯低于S組(P0.05或P0.01)。干預(yù)后第7天三組大鼠COX-2mRNA及蛋白表達(dá)、PGE2含量、caspase-3蛋白表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論肝部分切除術(shù)可導(dǎo)致海馬炎癥反應(yīng),帕瑞昔布可顯著抑制此種炎癥反應(yīng),其機(jī)制可能與下調(diào)海馬COX-2mRNA和蛋白表達(dá)、PGE2含量和caspase-3蛋白表達(dá)有關(guān)。
[Abstract]:Objective to study the effect of paroxib on hippocampal inflammation in elderly rats after partial hepatectomy. Methods 70 male Sprague-Dawley rats of SPF grade were randomly divided into three groups: control group C (n = 10), operation group S (n = 30) and paroxib group P (n = 30). Partial hepatectomy was used to establish the model of hippocampal inflammation induced by surgical trauma. Group P was subjected to partial hepatectomy under isoflurane anesthesia. The rats in group S were given partial hepatectomy under isoflurane anesthesia after intraperitoneal injection of paroxib (10 mg / kg) 1 h before operation and daily intraperitoneal injection of paroxib (2mg/kg) until the rats in group S were killed, and the rats in group S received partial hepatectomy under isoflurane anesthesia. Intraperitoneal injection of the same amount of saline as that of group P was performed 1 hour before operation. Group C was injected intraperitoneally with the same amount of saline as those in the above groups. The rats were killed on the 7th day after intervention. The hippocampal tissues were taken out and the expression of cyclooxygenase-2 and caspase-3 were detected by real-time quantitative PCR method. Western blot was used to detect the protein expression of COX-2 and cysteine protease -3caspase-3. The content of prostaglandin E2PGE2 was detected by Elisa. Results on the 1st day after intervention, the expression of COX-2mRNA and protein in group S and group P were significantly higher than those in group C (P0.05 or P0.01), but in group P was significantly lower than that in group S (P0.05 or P0.01). On the 7th day after intervention, there was no significant difference in the expression of COX-2mRNA and PGE2 and caspase-3 protein in the three groups. Conclusion partial hepatectomy can induce hippocampal inflammation, which can be significantly inhibited by paroxib, which may be related to the down-regulation of the expression of COX-2mRNA and protein in hippocampus and the expression of caspase-3 protein.
