【摘要】：目的：通過(guò)建立大鼠根尖周炎模型，研究糞腸球菌對(duì)大鼠根尖周炎NF-κB p65表達(dá)的影響。 方法：選取健康雄性SD大鼠24只，使用小號(hào)球鉆于右側(cè)下頜第一磨牙近中窩行開(kāi)髓術(shù)后，隨機(jī)分為A、B兩組，每組各12只。A組開(kāi)髓后，髓腔內(nèi)封入PBS緩沖液小棉球，設(shè)為未封菌組；B組開(kāi)髓后，髓腔內(nèi)封入糞腸球菌與PBS緩沖液調(diào)制的菌懸液小棉球，設(shè)為封菌組。兩組均使用進(jìn)口玻璃離子粘固劑封閉髓腔。左側(cè)下頜第一磨牙為空白對(duì)照組。常規(guī)條件飼養(yǎng)，A、B兩組分別于術(shù)后1周、2周、3周、4周四個(gè)時(shí)間點(diǎn)隨機(jī)各取3只鼠，水合氯醛麻醉處死后，分離其下頜骨，用小號(hào)挖匙挖取右側(cè)下頜第一磨牙和左側(cè)下頜第一磨牙根尖區(qū)牙槽骨、牙周膜組織，之后對(duì)組織進(jìn)行研磨，，提取、純化組織RNA，并逆轉(zhuǎn)錄成cDNA，采用實(shí)時(shí)熒光定量PCR (Real-time PCR)手段檢測(cè)各組NF-κB p65基因的表達(dá)水平并進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果： 1.健康空白對(duì)照組的NF-κB p65mRNA有少量表達(dá)，1周到4周A、B兩組各時(shí)間點(diǎn)的NF-κB p65mRNA的表達(dá)量均高于相應(yīng)空白對(duì)照組，差異有統(tǒng)計(jì)學(xué)意義。（P0.05） 2．A、B兩組術(shù)后1周到2周NF-κB p65mRNA的表達(dá)水平逐漸增高，2周表達(dá)水平達(dá)最高峰，3周表達(dá)水平開(kāi)始降低，4周表達(dá)水平繼續(xù)降低。四個(gè)實(shí)驗(yàn)組中1周與4周之間的差異無(wú)統(tǒng)計(jì)學(xué)意義（P0.05），其余各組之間的差異均具有統(tǒng)計(jì)學(xué)意義（P0.05）。 3．術(shù)后2周、3周，B組NF-κB p65mRNA的表達(dá)量均高于A組，差異均具有統(tǒng)計(jì)學(xué)意義（P0.05）。 結(jié)論： 1．NF-κB p65基因的表達(dá)水平可能與根尖周炎的發(fā)生、發(fā)展密切相關(guān)，提示NF-κB p65參與了根尖周炎的炎癥反應(yīng)和骨破壞。 2．NF-κB p65與糞腸球菌感染為主的根尖周炎密切相關(guān)，提示糞腸球菌存可能加重了根尖周組織的炎癥反應(yīng)和骨破壞。
[Abstract]:Aim: to study the effect of Enterococcus faecalis on the expression of NF- 魏 B p65 in rat periapical periodontitis. Methods: Twenty-four male Sprague-Dawley (SD) rats were randomly divided into two groups: group A (n = 12) and group A (n = 12). PBS buffer was injected into the medullary cavity of 24 male Sprague-Dawley rats. After opening the pulp in group B of unsealed bacteria, the bacteria suspension of Enterococcus faecalis and PBS buffer was added into the medullary cavity. Both groups were treated with imported glass ionomer to seal the medullary cavity. The left mandibular first molar was a blank control group. Three rats were randomly selected from each group at the time points of 1 week, 2 weeks, 3 weeks and 4 days after routine feeding. The mandible was isolated after anesthesia with chloral hydrate. The alveolar bone and periodontal ligament of the right first molar and the left first molar of the right mandible were extracted with a small spoon, and then the tissue was ground and extracted. The expression level of NF- 魏 B p65 gene in each group was detected by real-time fluorescence quantitative PCR real-time PCR, and the expression level of NF- 魏 B p65 gene was analyzed statistically. Results: 1. The expression of NF- 魏 B p65mRNA in the healthy blank control group was significantly higher than that in the control group at each time point from 1 to 4 weeks. The difference was statistically significant (P 0.05). 2. The expression level of NF- 魏 B p65mRNA increased gradually from 1 week to 2 weeks after operation in both groups. The expression level of NF- 魏 B increased gradually at the end of 2 weeks. The expression level of NF- 魏 B began to decrease at the end of 3 weeks and the expression level of NF- 魏 B decreased at the 4th week. There was no significant difference between one week and four weeks in the four experimental groups, but the differences among the other groups were statistically significant. 3. The expression of NF- 魏 B p65mRNA in group B was significantly higher than that in group A at 2 weeks and 3 weeks after operation, and the difference was statistically significant (P 0.05). Conclusion: 1. The expression level of NF- 魏 B p65 gene may be closely related to the occurrence and development of periapical inflammation, suggesting that NF- 魏 B p65 is involved in the inflammatory response and bone destruction in periapical periodontitis. 2. NF- 魏 B p65 was closely related to periapical periodontitis with Enterococcus faecalis infection, suggesting that the presence of Enterococcus faecalis may aggravate inflammation and bone destruction in periapical tissue.
