傳統(tǒng)藏藥“帕珠丸”對(duì)大鼠慢性乙醇性肝損傷的保護(hù)作用及其機(jī)制的研究

【摘要】:目的:研究傳統(tǒng)藏藥“帕珠丸”對(duì)慢性乙醇性肝。╝lcoholic liver disease,ALD)大鼠肝功能和組織酶活性的作用,以及對(duì)PPAR-α1和AMPK-α1基因表達(dá)水平的影響,,探討帕珠丸對(duì)慢性乙醇性肝損傷大鼠的保護(hù)作用及其可能的機(jī)制。 方法:將78只純系雄性SD大鼠適應(yīng)性喂養(yǎng)一周后,隨機(jī)分成正常對(duì)照組、模型對(duì)照組、聯(lián)苯雙酯組、帕珠丸低、中、高劑量組6組;模型對(duì)照組18只,其余每組各12只大鼠。模型組、聯(lián)苯雙酯組、帕珠丸低、中、高劑量組分別給予56%紅星二鍋頭酒10mL·kg-1灌胃;正常組給予10mL·kg-1生理鹽水灌胃,每天上午1次,連續(xù)灌胃12周,復(fù)制乙醇性肝損傷模型。造模成功后,帕珠丸低、中、高劑量組分別給予10mL·kg-1帕珠丸混懸液灌胃,劑量依次為0.05,0.10,0.20g·kg-1;聯(lián)苯雙酯組給予10mL·kg-1聯(lián)苯雙酯滴丸混懸液灌胃,劑量為0.003g·kg-1;正常對(duì)照組、模型對(duì)照組分別給予同體積生理鹽水灌胃,每天上午灌胃1次,連續(xù)灌胃3周。24h禁水禁食,麻醉,解剖,取樣。檢測(cè)血清丙氨酸轉(zhuǎn)氨酶(ALT)、天門冬氨酸轉(zhuǎn)氨酶(AST)、甘油三酯(TG)、超氧化物歧化酶(SOD)、還原性谷胱甘肽(GSH)、谷胱甘肽過氧化物酶(GSH-Px)、丙二醛(ADM)含量。并檢測(cè)肝組織中PPAR-α1mRNA、AMPK-α1mRNA的相對(duì)表達(dá)水平。 結(jié)果:與正常組比較,模型組大鼠肝臟濕重和肝指數(shù)和血清中ALT、AST、TG、MDA均明顯升高(P<0.05),SOD、GSH、GSH-Px均明顯降低(P<0.05),模型組大鼠肝組織中PPAR-α1mRNA、AMPK-α1mRNA表達(dá)量顯著降低(P<0.01);與模型組比較,帕珠丸各劑量組與聯(lián)苯雙酯組肝臟濕重和肝指數(shù)和血清中ALT、AST、TG均明顯降低(P<0.05),MDA顯著降低(P<0.01),SOD、GSH、GSH-Px均明顯升高(P<0.05);與模型組比較,帕珠丸各劑量組與聯(lián)苯雙酯組PPAR-α1mRNA、AMPK-α1mRNA表達(dá)量均顯著升高(P<0.05)。結(jié)論:帕珠丸能有效保護(hù)乙醇對(duì)大鼠肝臟的損害,改善肝功能,降低甘油三酯水平,抑制ROS引起的脂質(zhì)過氧化反應(yīng)。提示該藥對(duì)大鼠乙醇性肝損傷有較好的保護(hù)作用。
[Abstract]:Objective: to study the effects of traditional Tibetan medicine Pazhuwan on liver function and tissue enzyme activity in rats with chronic alcoholic liver disease (liver) and the expression level of PPAR- 偽 1 and AMPK- 偽 1 genes.To investigate the protective effect of Pazhuwan on chronic ethanol induced liver injury in rats and its possible mechanism.Methods: 78 male Sprague-Dawley rats were randomly divided into normal control group, model control group, biphenyl diester group, pazhu pill low, middle and high dose group, and model control group with 18 rats, 12 rats in each group.Model group, Biphenyl diester group, Pazhuwan low, medium and high dose group were given 56% Hongxing Erguotou 10mL kg-1, and normal group were given 10mL kg-1 saline once a day for 12 weeks, to make ethanol induced liver injury model.After the model was successfully made, Pazu pills were given 10mL kg-1 pellet suspension by stomach in low, medium and high dosage groups, the dosage was 0.05 ~ 0.100.100.20g / kg -1 respectively, the biphenyl diester group was given 10mL kg-1 biphenyl diester dropping pill suspension, the dosage was 0.003g / kg ~ (-1), the normal control group, the control group, the control group, the control group, the control group, the control group, the control group, the control group and the control group, respectively.The model control group was fed with the same volume of normal saline once a day for 3 weeks, 24 hours, anaesthetized, dissected and sampled.Serum alanine aminotransferase (alt), aspartate aminotransferase (AST), triglyceride (TG), superoxide dismutase (SOD), reduced glutathione (GSH), glutathione peroxidase (GSH-PxN), malondialdehyde (MDA) were measured.The relative expression of PPAR- 偽 1 mRNA-AMPK- 偽 1mRNA in liver tissue was detected.緇撴灉:涓庢甯哥粍姣旇緝,妯″瀷緇勫ぇ榧犺倽鑴忔箍閲嶅拰鑲濇寚鏁板拰琛
