大鼠腦損傷后血漿ET-1的含量變化與損傷時(shí)間的關(guān)系

【摘要】:目的:應(yīng)用酶聯(lián)免疫法、放射免疫法定量測(cè)定SD大鼠腦損傷后血漿ET-1的表達(dá)規(guī)律。尋找ET-1的表達(dá)與損傷時(shí)間的關(guān)系,為法醫(yī)學(xué)腦損傷的時(shí)間推斷提供更為完善的方法。 方法:選取健康成年SD大鼠64只,雌雄不限,隨機(jī)分為正常組(n=8),假損傷對(duì)照組(n=8)以及損傷實(shí)驗(yàn)組(n=48),實(shí)驗(yàn)組又按損傷時(shí)間不同分為6個(gè)亞組。實(shí)驗(yàn)組大鼠實(shí)施麻醉后,參照Feeney s法,采用自由落體打擊裝置,,于顱骨正中線旁3mm處造成腦損傷模型。于損傷后1h、6h、24h、48h、3d、7d六個(gè)時(shí)間點(diǎn)(n=8)采血4ml及2ml,離心分離血漿后置-20℃冰箱保存。正常組不做任何處理,假損傷對(duì)照組僅做鉆孔處理,不做自由落體打擊,不致腦損傷。應(yīng)用酶聯(lián)免疫法以及放射免疫法對(duì)大鼠腦損傷后血漿ET-1進(jìn)行定量檢測(cè),并對(duì)檢測(cè)結(jié)果進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:(1)ELISA法:大鼠腦損傷后血漿ET-1含量于傷后1h開(kāi)始升高(P0.05),與損傷后24h達(dá)到高峰。損傷后48h開(kāi)始逐漸下降,3d后開(kāi)始逐漸趨于假損傷組水平(P0.05),7d后基本恢復(fù)正常值。(2)RIA法:大鼠腦損傷后血漿ET-1含量表達(dá)規(guī)律基本與ELISA法所測(cè)結(jié)果表達(dá)規(guī)律一致。 結(jié)論:大鼠腦損傷后血漿ET-1的含量隨損傷時(shí)間呈規(guī)律性變化。兩種檢測(cè)方法所得結(jié)果呈一致性。酶聯(lián)免疫法及放射免疫法操作簡(jiǎn)單易行,可作為日后法醫(yī)學(xué)腦損傷鑒定及研究的依據(jù)。
[Abstract]:Objective: to investigate the relationship between the expression of ET-1 and the time of brain injury in SD rats by enzyme linked immunosorbent assay (Elisa) and radioimmunoassay (RIA), and to provide a more perfect method for estimating the time of brain injury in forensic medicine. Methods: 64 healthy adult SD rats, male and female, were randomly divided into normal group (n = 8) and sham injury control group (n = 48). The experimental group was also divided into 6 subgroups according to the time of injury. The experimental group was anesthetized. According to Feeney s method, the model of brain injury was made at 3mm near the midline of skull by using free fall device. The blood samples were collected at 6 h, 6 h, 24 h, 48 h, 3 d, 7 d, six time points, 6 ml and 2 ml, then centrifuged plasma was stored in refrigerator at -20 鈩
