【摘要】：目的探討p38MAPK信號(hào)通路在地佐辛減輕大鼠切口痛中的作用。方法雄性SD大鼠32只,體重250~300g,隨機(jī)均分為四組:假手術(shù)組(S組)、切口痛組(I組)、地佐辛0.1mg/kg預(yù)處理組(DL組)和地佐辛0.2mg/kg預(yù)處理組(DH組)。S組:僅給予麻醉;I組、DL組、DH組建立切口痛模型,其中DL組和DH組在建立模型前30min通過(guò)腹腔注射0.1mg/kg和0.2mg/kg的地佐辛,I組建立模型前30min腹腔注射同體積的生理鹽水。按Brennan法制成大鼠切口痛模型,以von Frey細(xì)絲法、熱輻射法觀察大鼠術(shù)前30min、術(shù)后4、8、24h機(jī)械刺激縮足反應(yīng)閾值(MWT)和輻射熱刺激縮足反應(yīng)潛伏期(WTL)。術(shù)后24h取相應(yīng)節(jié)段脊髓,并以Western blot法檢測(cè)脊髓pp38MAPK蛋白表達(dá)。結(jié)果與S組比較,I、DL和DH組術(shù)后4、8、24h的MWT明顯降低和WTL明顯縮短(P0.05);與I組比較,DL和DH組MWT明顯增高和WTL均明顯延長(zhǎng)(P0.05);與DL組比較,DH組MWT增高和WTL延長(zhǎng),但組間差異無(wú)統(tǒng)計(jì)學(xué)意義。與S組比較,術(shù)后24hI、DL和DH組脊髓p-p38MAPK蛋白表達(dá)明顯增加(P0.05);與I組比較,DL和DH組脊髓pp38MAPK蛋白表達(dá)明顯降低(P0.05);與DL組比較,DH組p-p38MAPK蛋白表達(dá)降低,但組間差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論術(shù)前腹腔注射地佐辛的抗傷害作用可能與抑制脊髓p-p38MAPK表達(dá)有關(guān)。
[Abstract]:Objective to investigate the role of p38 MAPK signaling pathway in relieving incision pain in rats with dizosin. Methods 32 male Sprague-Dawley rats were used. The rats were randomly divided into four groups: sham operation group (group S), incision pain group (group I), dizosin 0.1 mg / kg preconditioning group (group D) and dizosin group (0.2 mg / kg preconditioning group) and dh group (group S). In DL group and DH group, intraperitoneal injection of 0.1 mg / kg and 0.2 mg / kg of dizosinone I was used to establish the model 30 minutes before the model was established by intraperitoneal injection of normal saline of the same volume. The rat model of incision pain was established by Brennan method and von Frey filament method was used. The thermal radiation method was used to observe the threshold value of mechanical stimulation of foot contraction response (MWTs) and the latency of radiation heat stimulation of foot contraction reaction in rats 30 minutes before operation and 8 minutes after operation for 24 hours. The corresponding segments of spinal cord were taken out 24 hours after operation, and the corresponding segments of spinal cord were taken out at 24 hours after operation. The expression of pp38MAPK protein in spinal cord was detected by Western blot method. Results compared with group S and group DH, MWT decreased significantly and WTL shortened significantly at 8h after operation (P 0.05), MWT in group I and DH were significantly increased and WTL was significantly prolonged compared with group DL, and DH was significantly longer than that in group DL. MWT increased and WTL prolonged. Compared with group S, the expression of p-p38 MAPK protein in spinal cord increased significantly in IDL and DH groups at 24 h after operation, the expression of p-p38 MAPK protein in spinal cord in group I and group I decreased significantly compared with group I, and the expression of p-p38 MAPK protein in group D was significantly lower than that in group D (P < 0.05), and the expression of p-p38 MAPK protein in group D was significantly lower than that in group D (P < 0.05). Conclusion the anti-nociceptive effect of intraperitoneal injection of dizosin may be related to the inhibition of p-p38 MAPK expression in the spinal cord.
