中藥仙茅對(duì)大鼠腓腸肌單收縮張力及mTOR激活效應(yīng)影響的研究

【摘要】:1.研究目的 通過對(duì)灌服中藥仙茅制劑后的SD大鼠不同時(shí)段的腓腸肌單收縮張力、肌組織IGF-1含量和雷帕霉素靶蛋白(mTOR)mRNA的表達(dá)水平的測(cè)定,探討中藥仙茅對(duì)增強(qiáng)骨骼肌收縮能力的機(jī)制和時(shí)間效應(yīng)。 2.研究方法 以9周齡SD雄性大鼠為實(shí)驗(yàn)研究對(duì)象,隨機(jī)分為4組,即空白組,灌胃后90min組,灌胃后120min組,灌胃后240min組,每組8只,分籠飼養(yǎng)。經(jīng)過一周的常規(guī)飼養(yǎng)后,對(duì)其進(jìn)行為期2天的灌胃適應(yīng),灌注蒸餾水。在第3天,經(jīng)過12h的禁食禁水之后,對(duì)除空白組之外的三組大鼠灌胃中藥仙茅制劑,而空白組灌胃等量的蒸餾水。分別在灌胃后90、120和240min對(duì)大鼠進(jìn)行乙醚麻醉后,游離一側(cè)腓腸肌遠(yuǎn)端,借助BL-420F生物信號(hào)采集處理系統(tǒng)測(cè)試并記錄大鼠腓腸肌單收縮張力,,并取大鼠另一側(cè)腓腸肌入液氮速凍,之后分別采用elisa法和RT-PCR法檢測(cè)大鼠腓腸肌IGF-1的含量和mTORmRNA的表達(dá)。 3.實(shí)驗(yàn)結(jié)果 3.1與對(duì)照組相比,灌藥的3組大鼠腓腸肌單收縮張力較空白組顯著升高(P0.05),且240min組較其他幾組升高最為顯著。 3.2灌胃后IGF-1的含量隨時(shí)間延長(zhǎng)顯著上升,120min組顯著高于90min組(P0.05),240min組極顯著高于120min組(P0.01),且灌服中藥的3組均明顯高于空白組(P0.05)。 3.3空白組的大鼠腓腸肌mTORmRNA表達(dá)量顯著低于灌藥的3組,且240min組顯著高于120min組(P0.05),120min組顯著高于90min組(P0.05),由90min組到240min組隨著時(shí)間的推移,mTORmRNA的表達(dá)量也呈現(xiàn)逐漸升高的趨勢(shì)。 4.研究結(jié)論: 4.1中藥仙茅可明顯提高大鼠腓腸肌單收縮張力。 4.2中藥仙茅可明顯提高大鼠腓腸肌IGF-1的含量,并能促進(jìn)mTORmRNA的表達(dá),且在服藥后的前4個(gè)小時(shí)之內(nèi)表達(dá)量逐漸上升。 4.3服藥后IGF-1的含量上升,mTORmRNA的表達(dá)量也出現(xiàn)上升,提示服用仙茅確實(shí)對(duì)骨骼肌蛋白合成具有一定的促進(jìn)作用。
[Abstract]:1. Purpose of the study. The contractile tension of gastrocnemius muscle, the content of IGF-1 in muscle tissue and the expression level of rapamycin target protein mTORN mRNA in gastrocnemius muscle of SD rats were measured after administration of Chinese herbal medicine. Objective: to study the mechanism and time effect of Chinese herbal medicine Festuca arundinacea on enhancing skeletal muscle contraction. 2. Research methodology. Sprague-Dawley (SD) male rats aged 9 weeks were randomly divided into four groups: blank group, 90 min group, 120min group and 240 min group. On the third day, after 12 hours of fasting and water ban, three groups of rats, except blank group, were treated with Chinese herbal medicine Festuca arundinacea preparation. In blank group, rats were anesthetized with ether 90,120 and 240 minutes after administration of distilled water, and the contractile tension of gastrocnemius muscle was measured and recorded by BL-420F biological signal acquisition and processing system. The other side of gastrocnemius muscle was frozen with liquid nitrogen, and then the content of IGF-1 and the expression of mTORmRNA in gastrocnemius muscle were detected by elisa and RT-PCR methods respectively. 3. Experimental results. 3.1 compared with the control group, the contractile tension of gastrocnemius in the three groups was significantly higher than that in the blank group, and that in the 240min group was significantly higher than that in the other groups. The content of IGF-1 in 120min group was significantly higher than that in 90min group (P 0.05A 240min group), which was significantly higher than that in 120min group (P 0.01), and the level of IGF-1 in the three groups was significantly higher than that in the blank group (P 0.05). 3.3 the expression of mTORmRNA in gastrocnemius muscle in blank group was significantly lower than that in the control group, and was significantly higher in 240min group than that in 120min group, which was significantly higher than that in 90min group and 90min group. The expression of mTOR mRNA in the group from 90min to 240min showed a trend of increasing with the passage of time. 4. Conclusions of the study:. 4.1 the single contractile tension of gastrocnemius muscle in rats was significantly increased by Festuca arundinacea. 4.2 the content of IGF-1 in gastrocnemius muscle of rats was obviously increased, and the expression of mTORmRNA was increased gradually in the first 4 hours after administration of Festuca arundinacea. 4.3 the content of IGF-1 increased and the expression of mTO mRNA increased, suggesting that citronella did promote skeletal muscle protein synthesis.
