L-肉堿對(duì)NAFLD脂肪肝大鼠肝臟線粒體能量代謝及抗氧化能力的影響

【摘要】:研究L-肉堿對(duì)NAFLD脂肪肝大鼠肝臟線粒體能量代謝及抗氧化能力的影響。脂肪肝大鼠48只,隨機(jī)分為對(duì)照組、肉堿組、運(yùn)訓(xùn)組和肉堿+運(yùn)訓(xùn)組。運(yùn)訓(xùn)組和肉堿+運(yùn)訓(xùn)組進(jìn)行6周運(yùn)動(dòng),肉堿組和肉堿+運(yùn)訓(xùn)組每日灌胃一次肉堿(500 mg/kg)。6周后各組大鼠進(jìn)行力竭運(yùn)動(dòng),即刻麻醉取出肝臟提取線粒體,測(cè)定線粒體呼吸鏈酶RCCI~IV活性及自由基代謝相關(guān)指標(biāo)。與對(duì)照組比,肉堿組大鼠肝臟線粒體呼吸鏈RCCII,運(yùn)訓(xùn)組RCCI、RCCIII,肉堿+運(yùn)訓(xùn)組RCCI、RCCII、RCCIII活性均顯著提高。與肉堿組比,肉堿+運(yùn)訓(xùn)組RCCI、RCCIII活性顯著提高。與運(yùn)訓(xùn)組比,肉堿+運(yùn)訓(xùn)組RCCI活性顯著提高。與對(duì)照組比,肉堿組、肉堿+運(yùn)訓(xùn)組大鼠肝臟線粒體GSH-Px活性顯著提高;與對(duì)照組比,三組SOD活性顯著提高,MDA含量降低。與肉堿組比,肉堿+運(yùn)訓(xùn)組SOD活性顯著提高;與運(yùn)訓(xùn)組比,肉堿+運(yùn)訓(xùn)組GSH-Px活性顯著提高。補(bǔ)充L-肉堿、運(yùn)動(dòng)、L-肉堿+運(yùn)動(dòng)均可提高非酒精性脂肪肝大鼠肝臟線粒呼吸鏈酶活性,提高線粒體能量代謝速率及抗氧化損傷能力,補(bǔ)充L-肉堿結(jié)合運(yùn)動(dòng)訓(xùn)練組效果更加顯著。
[Abstract]:To study the effects of L-carnitine on energy metabolism and antioxidant capacity of mitochondria in NAFLD fatty liver rats, 48 rats with fatty liver were randomly divided into control group and carnitine group. Training group and carnitine training group were trained for 6 weeks, carnitine training group and carnitine training group were fed with carnitine once a day for 500 mg/kg).6 weeks, the rats in each group were subjected to exhaustive exercise, and liver mitochondria were extracted immediately after anesthesia. The activity of mitochondrial respiratory chain enzyme (RCCI~IV) and free radical metabolism in carnitine group was significantly higher than that in carnitine group, RCCIII activity in carnitine group, RCCIII activity in carnitine training group, RCCIII activity in carnitine training group, and in carnitine training group. Compared with the training group, the activity of RCCI in the carnitine training group and the carnitine group, the activity of mitochondrial GSH-Px in the liver of the carnitine training group and the carnitine training group were significantly higher than those in the control group and the control group, respectively, and compared with the control group, the activity of GSH-Px in the liver of the carnitine training group was significantly higher than that in the carnitine training group. Compared with carnitine group, SOD activity of carnitine training group increased significantly, and that of carnitine training group significantly increased compared with carnitine training group. L- carnitine exercise could increase the activity of linear respiratory chain enzyme in liver of non-alcoholic fatty liver rats, increase the rate of energy metabolism of mitochondria and the ability of antioxidant damage, and the effect of supplementation of L-carnitine combined with exercise training group was more remarkable.
