新型等離子體射流對(duì)糞腸球菌的滅菌實(shí)驗(yàn)研究

【摘要】:目的:本研究旨在探索一種全新的、有效的根管消毒方法,提高根管消毒的效能,從而提高根管治療術(shù)的成功率。 方法:本研究采用一種新型納秒脈沖驅(qū)動(dòng)的大氣壓非平衡等離子體射流分別作用于糞腸球菌(Enterococcus faecalis,E. faecalis)標(biāo)準(zhǔn)株培養(yǎng)皿、E. faecalis硝酸纖維素生物膜模型以及離體牙根管E. faecalis感染模型。該新型等離子體射流在各物理參數(shù)恒定情況下,分別以不同時(shí)間、不同環(huán)境及不同形式作用于培養(yǎng)皿、生物膜模型和離體牙感染根管模型,對(duì)E. faecalis的滅菌效果進(jìn)行了觀察和測(cè)定,并分析了該等離子體射流滅菌效果的影響因素,還通過(guò)透射電鏡觀察處理后細(xì)菌形態(tài)的變化分析其滅菌機(jī)理。同時(shí)對(duì)經(jīng)Micro-CT掃描觀察到的各離體牙的根管形態(tài)與滅菌效果之間的關(guān)系進(jìn)行了分析。 結(jié)果:本研究結(jié)果顯示此新型大氣壓非平衡等離子體射流對(duì)體外培養(yǎng)的E. faecalis有良好的滅菌效果,并對(duì)E. faecalis生物膜模型及離體牙感染E. faecalis模型有一定的消毒滅菌作用。但這些滅菌效果與等離子體射流作用時(shí)間、作用環(huán)境以及感染根管的形態(tài)密切相關(guān)。 結(jié)論:此新型大氣壓非平衡等離子體射流可以作為根管消毒的輔助方法,但其滅菌作用的可靠性、有效性、安全性及其作用機(jī)理仍需進(jìn)一步實(shí)驗(yàn)研究證實(shí)。
[Abstract]:Objective: to explore a new and effective method of root canal disinfection to improve the efficacy of root canal disinfection and to improve the success rate of root canal therapy. Methods: in this study, a novel nanosecond pulse-driven atmospheric pressure non-equilibrium plasma jet was applied to Enterococcus faecalis E. faecalis standard strain, E. faecalis nitrate cellulose biofilm model and in vitro root canal E. faecalis infection model. The new plasma jet acted on petri dish, biofilm model and tooth infected root canal model in different time, environment and form under constant physical parameters. The sterilizing effect of E. faecalis was observed and measured, and the influencing factors of the effect of plasma jet sterilization were analyzed. The sterilizing mechanism was also analyzed by observing the morphological changes of bacteria after treatment with transmission electron microscope. At the same time, the relationship between the root canal morphology and sterilizing effect of the isolated teeth observed by Micro-CT scan was analyzed. Results: the results showed that the new atmospheric pressure non-equilibrium plasma jet had a good sterilizing effect on E. faecalis cultured in vitro, and had a certain disinfection and sterilization effect on E. faecalis biofilm model and the model of isolated tooth infection E. faecalis. However, these sterilization effects are closely related to the time of plasma jet action, the environment of action and the morphology of infected root canal. Conclusion: this new type of atmospheric pressure non-equilibrium plasma jet can be used as an auxiliary method for root canal disinfection, but the reliability, effectiveness, safety and mechanism of its sterilization need further experimental study.
