狂犬病疫苗與蟾皮提取液聯(lián)合應(yīng)用預(yù)防狂犬病效果的研究

【摘要】: 蟾皮作為傳統(tǒng)中藥的重要藥材,在咳喘、肝硬化腹水、濕疹、腫瘤、疳積、咽喉腫痛、癰腫等諸多病癥的治療中起舉足輕重的作用。在這些疾病的治療中蟾皮的免疫學(xué)作用日益體現(xiàn)出來,但由于現(xiàn)有蟾皮制劑有一定得副作用,不能滿足用于免疫治療制劑對(duì)安全性的要求,未在免疫學(xué)領(lǐng)域廣泛應(yīng)用。本試驗(yàn)對(duì)蟾皮提取液進(jìn)行了初步純化,去除刺激性較大的成分以提高其安全性。狂犬病作為目前我國(guó)傳染病疫情中造成死亡人數(shù)最多的傳染病,目前還沒有找到有效的治療方法。暴露后以狂犬病疫苗免疫是主要的預(yù)防手段,能否產(chǎn)生高效價(jià)的中和性抗體和良好的細(xì)胞免疫對(duì)狂犬病的防治起到至關(guān)重要的作用。 本文進(jìn)一步純化蟾皮提取液,并與狂犬病疫苗在動(dòng)物中聯(lián)合應(yīng)用,觀察其細(xì)胞免疫效果,指標(biāo)有脾淋巴細(xì)胞轉(zhuǎn)化率,CD4+、CD4+/CD8+,巨嗜細(xì)胞吞噬率和吞噬指數(shù)。同時(shí)觀察其體液免疫效果,測(cè)定不同時(shí)間的中和抗體效價(jià)。經(jīng)統(tǒng)計(jì)學(xué)分析狂犬病疫苗與蟾皮提取物聯(lián)合應(yīng)用具有明顯增強(qiáng)機(jī)體細(xì)胞免疫的功能,能夠快速產(chǎn)生抗病毒抗體,對(duì)主動(dòng)的細(xì)胞免疫有很好的協(xié)同作用。
[Abstract]:As an important traditional Chinese medicine, Xenopus skin plays an important role in the treatment of cough and asthma, cirrhotic ascites, eczema, tumor, infantile, throat swelling, carbuncle and so on. The immunological effect of Xenopus skin is increasingly reflected in the treatment of these diseases, but the existing Xenopus skin preparations have some side effects, which can not meet the safety requirements of immunotherapy agents, and have not been widely used in immunological field. In order to improve the safety of Xenopus skin extract, the extract of Xenopus skin was preliminarily purified and the irritant components were removed. Rabies is one of the most common infectious diseases in China. Immunization with rabies vaccine after exposure is the main preventive method. It is very important to produce high titer neutralizing antibody and good cellular immunity for the prevention and treatment of rabies. In this paper, the extract of Xenopus skin was further purified and used in combination with rabies vaccine in animals to observe its cellular immune effect. The indexes included spleen lymphocyte transformation rate, CD4 / CD8, phagocytosis rate and phagocytic index of giant eosinophils. At the same time, the humoral immune effect was observed, and the neutralizing antibody titer at different time was determined. The combination of rabies vaccine and Xenopus skin extract can obviously enhance the function of cellular immunity and produce anti-virus antibody quickly. It has a good synergistic effect on active cellular immunity.
