【摘要】：目的研究GFP標(biāo)記的復(fù)制缺陷型腺病毒在消毒劑滅活病毒試驗(yàn)中應(yīng)用的可行性。方法采用商品化Ad-easy系統(tǒng)經(jīng)同源重組包裝GFP標(biāo)記的復(fù)制缺陷型腺病毒,擴(kuò)增后進(jìn)行熒光計(jì)數(shù)和復(fù)制缺陷性能檢測,其作為指示病毒進(jìn)行病毒滅活試驗(yàn)。結(jié)果腺病毒基因同源重組后轉(zhuǎn)染AD-293細(xì)胞出現(xiàn)綠色熒光和細(xì)胞病變,成功包裝出病毒顆粒,擴(kuò)增后經(jīng)熒光計(jì)數(shù)病毒滴度為4.20×10~6gfu/ml。盲傳3代后,感染Vero細(xì)胞未出現(xiàn)熒光和細(xì)胞病變,表明無子代病毒的產(chǎn)生。以有效氯含量200 mg/L消毒劑作用于腺病毒20 min后,殺滅對數(shù)值為4.10。結(jié)論熒光標(biāo)記的復(fù)制缺陷型腺病毒在含氯消毒劑對病毒殺滅能力檢測評價(jià)中具有一定的可行性,更為高效和安全。
[Abstract]:Objective to study the feasibility of replication-deficient adenovirus labeled with GFP in disinfectant inactivation test. Methods Replication-deficient adenovirus labeled by GFP was packaged with commercial Ad-easy system. The results showed that the adenovirus gene homologous recombination showed green fluorescence and cytopathic changes in AD-293 cells, and the virus particles were successfully packaged. After amplification, the virus titer was 4.20 脳 10 ~ (-1) 6g / ml. After 3 generations of blind transmission, there was no fluorescence and cytopathic changes in infected Vero cells, indicating that no offspring virus was produced. The virus was treated with 200 mg/L disinfectant with available chlorine content for 20 min. Conclusion the fluorescent labeled replication-deficient adenovirus is feasible and more efficient and safe in the detection and evaluation of chlorinated disinfectant's ability to kill the virus.
