【摘要】：目的:體外培養(yǎng)大鼠下頜下腺細胞,并對其生物學(xué)特性進行研究,為涎腺疾病的探索提供體外模型。方法:無菌條件下取7d齡Wistar大鼠雙側(cè)下頜下腺,仔細分離脂肪、包膜、神經(jīng)和血管,采用組織塊培養(yǎng)法進行培養(yǎng)。運用酶消化法和差速貼壁法純化細胞;免疫組化SP法檢測Cytokeratin-8(CK-8)抗體和α-Amylase抗體的表達;PAS染色法檢測細胞糖原分泌;掃描電鏡(scanning electronic microscopy,SEM)觀察細胞的形態(tài)學(xué)特征。結(jié)果:組織塊培養(yǎng)法可成功獲得下頜下腺細胞,免疫組化染色可見大部分細胞胞質(zhì)為棕黃色,提示CK-8抗體表達陽性,α-Amylase抗體表達陽性;PAS染色可見胞質(zhì)呈紫紅色;SEM可見細胞伸出長短不一的偽足,胞核著色深,有些細胞可見分泌顆粒。結(jié)論:組織塊培養(yǎng)法可以成功培養(yǎng)大鼠下頜下腺細胞,并且操作簡便。
[Abstract]:Objective: To study the biological characteristics of submandibular gland cells in vitro, and to provide an in vitro model for the exploration of salivary gland diseases. Methods: The submandibular gland of 7 days old Wistar rats was taken under the condition of sterility, and the fat, the envelope, the nerves and the blood vessels were carefully separated, and the culture was carried out by the tissue culture method. The cells were purified by an enzyme digestion method and a differential adhesion method. The expression of the antibody of Cytokeratin-8 (CK-8) and the antigen-Aylase was detected by SP method. The cellular glycogen secretion was detected by PAS staining. The morphological characteristics of the cells were observed by scanning electron microscopy (SEM). Results: The submandibular gland cells were successfully obtained by the tissue culture method. The cytoplasm of most of the cells was brown yellow, and the expression of CK-8 was positive, and the expression of the antigen-Aylase was positive. PAS staining showed that the cytoplasm was purplish red, and the SEM showed that the cells extended out of the pseudo-foot with different lengths. The nuclei are colored deep and some cells are visible in the secretory granules. Conclusion: The submandibular gland cells of the rat can be cultured successfully by the tissue culture method, and the operation is simple and convenient.
