【摘要】：目的觀察二十二碳六烯酸(DHA)對(duì)牙齦卟啉單胞菌(P.gingivalis)脂多糖(LPS)誘導(dǎo)人牙周膜成纖維細(xì)胞(h PDLCs)表達(dá)炎癥因子的影響。方法體外培養(yǎng)鑒定h PDLCs,采用MTT法觀察不同濃度DHA對(duì)h PDLCs細(xì)胞活性的影響。根據(jù)MTT法的結(jié)果,選擇對(duì)細(xì)胞活性沒有影響的刺激濃度,分別采用real-time PCR和ELISA技術(shù)從基因和蛋白水平檢測(cè)DHA對(duì)P.gingivalis LPS誘導(dǎo)h PDLCs表達(dá)炎癥因子白細(xì)胞介素-6(IL-6)、IL-8和IL-1β的影響。結(jié)果在25~100μmol/L濃度范圍內(nèi),DHA對(duì)h PDLCs細(xì)胞活性沒有影響,且可呈濃度依賴性下調(diào)P.gingivalis LPS誘導(dǎo)h PDLCs表達(dá)的炎癥因子。結(jié)論在不影響細(xì)胞活性的情況下,DHA可在一定濃度范圍內(nèi),呈濃度依賴性下調(diào)P.gingivalis LPS誘導(dǎo)h PDLCs表達(dá)的炎癥因子,表明DHA具有用于牙周炎抗炎治療的可能性。
[Abstract]:Aim to investigate the effect of 22-carbohexaenoic acid (DHA) on (h PDLCs) expression in human periodontal ligament fibroblasts induced by lipopolysaccharide (LPS) of Porphyromonas gingivalis (P.gingivalis). Methods the effect of different concentrations of DHA on the activity of h-PDLCs cells was observed by MTT assay. H-PDLCs, was cultured and identified in vitro. According to the results of MTT assay, the stimulation concentration which had no effect on cell activity was selected. The expression of interleukin-6 (IL-6) in h-PDLCs induced by P.gingivalis LPS was detected by real-time PCR and ELISA at gene and protein levels, respectively. The effect of IL-8 and IL-1 尾. Results in the concentration range of 25 ~ 100 渭 mol / L, DHA had no effect on the activity of h-PDLCs cells, and P.gingivalis LPS-induced inflammatory cytokines induced h-PDLCs expression were down-regulated in a concentration-dependent manner. Conclusion DHA can down-regulate the expression of P.gingivalis LPS-induced h-PDLCs inflammatory factors in a concentration-dependent manner in a certain concentration range without affecting cell activity, suggesting that DHA may be used in anti-inflammatory therapy of periodontitis.
