具核梭桿菌對(duì)牙齦卟啉單胞菌或伴放線聚集桿菌誘導(dǎo)多形核白細(xì)胞活性氧生成的影響

【摘要】:目的:探討與具核梭桿菌(F.nucleatum,Fn)共同感染時(shí),牙齦卟啉單胞菌(P.gingivalis,Pg)或伴放線聚集桿菌(A.actinomycetemcomitans,Aa)對(duì)多形核白細(xì)胞(PMNs)產(chǎn)生活性氧(ROS)的影響。方法:取健康人抗凝外周血,經(jīng)Percoll分離后取PMNs,將分離的PMNs與2’,7’-二氯熒光素二乙酸酯(DCFH-DA)混合,于37℃的水浴箱中孵育15min,然后分別向管中加入A組和B組細(xì)菌(細(xì)菌分組:A1,Pg;A2,Aa;A3,Fn;B1,Pg+Fn;B2,Aa+Fn;B3,Pg+Aa;B4,Pg+Aa+Fn;B5,Aa+Pg+Fn),充分混合后于37℃孵育1h和4h,流式細(xì)胞術(shù)(Flow Cytometry,FCM)檢測PMNs產(chǎn)生ROS的量。結(jié)果:不同牙周致病菌與分離的PMNs在37℃作用1h,PMNs生成ROS的量分別是A1-16.52;A2-18.74;A3-19.59;B1-17.60;B2-20.09,其中B1A1;B2A2,差異顯著(P0.05)。不同微生物與分離的PMNs在37℃作用4h,PMNs生成ROS的量分別是A1-176.05;A2-203.19;A3-230.36;B1-259.08;B2-274.89,其中B1A1;B2A2,差異顯著(P0.05)。結(jié)論:與Fn共聚后,Pg或Aa誘導(dǎo)PMNs產(chǎn)生ROS的能力被增強(qiáng)。
