內毒素耐受對人中性粒細胞抗炎反應的影響

【摘要】:目的:探討脂多糖(lipopolysaccharide,LPS)誘導的耐受對人中性粒細胞分泌抗炎因子白介素(interleukin,IL)-10及細胞凋亡水平的影響。方法:采用1μg/m L牙齦卟啉單胞菌(Porphyromonasgingivalis,P.gingivalis)LPS或者1μg/m L大腸桿菌(Escherichia coli,E.coli)LPS刺激人中性粒細胞12 h,誘導細胞耐受。洗脫后,分別采用2種LPS再次刺激20 h,采用ELISA技術檢測細胞分泌IL-10水平。同法誘導細胞耐受,洗脫后,再次刺激3 h,采用AV-PI法檢測細胞凋亡水平。結果:P.gingivalis LPS或E.coli LPS單次刺激人中性粒細胞后,IL-10分泌水平均較刺激前明顯增加(P0.05),凋亡水平則無明顯改變(P0.05)。2種LPS分別重復刺激之后,IL-10分泌水平均較單次刺激后顯著增加(P0.05)。P.gingivalis LPS重復刺激3 h后,中性粒細胞短期內凋亡水平與單次刺激后無明顯改變(P0.05),而E.coli LPS重復刺激3 h后,細胞短期內凋亡水平較單次刺激后明顯下降(P0.05)。結論:內毒素耐受能促進人中性粒細胞分泌IL-10,但不同LPS誘導的耐受對細胞凋亡水平的影響可能存在差異。
[Abstract]:Aim: to investigate the effects of lipopolysaccharide (lipopolysaccharide,LPS) -induced tolerance on the secretion of anti-inflammatory factor interleukin-10 (interleukin,IL) and apoptosis by human neutrophil. Methods: 1 渭 g / mL porphyromonas gingivalis (Porphyromonasgingivalis,P.gingivalis) LPS or 1 渭 g / mL Escherichia coli (Escherichia coli,E.coli) LPS were used to induce human neutrophil tolerance for 12 h. After elution, two kinds of LPS were used to stimulate again for 20 h, and ELISA technique was used to detect the level of IL-10 secreted by cells. Cell tolerance was induced by the same method. After elution, the level of apoptosis was detected by AV-PI assay. Results: after stimulation of human neutrophils with P.gingivalis LPS or E.coli LPS, the level of IL-10 secretion increased significantly (P0.05), while the level of apoptosis did not change (P0.05). The level of IL-10 secretion was significantly higher than that of single stimulation (P0.05). After 3 h of repeated P.gingivalis LPS stimulation, the level of neutrophil apoptosis in the short term was not significantly changed compared with that after single stimulation (P0.05). After repeated stimulation of E.coli LPS for 3 h, the level of apoptosis in a short period of time was significantly lower than that after a single stimulation (P0.05). Conclusion: endotoxin tolerance can promote the secretion of IL-10, by human neutrophils. However, the effects of different LPS induced tolerance on apoptosis may be different.
