非平衡等離子體對感染根管細菌生物膜影響的實驗研究

【摘要】:目的:大量研究資料表明,非平衡等離子體具有良好的滅菌效果。由于其氣體溫度接近室溫,對機體組織無明顯損害,活性成分存活周期短而無毒性物質(zhì)殘留,能夠到達微小不規(guī)則的部位,其在醫(yī)學領(lǐng)域的應(yīng)用受到了越來越多的關(guān)注。本課題組的早期實驗研究證實,非平衡等離子體能夠有效地殺滅糞腸球菌生物膜,其滅菌效果與常規(guī)根管沖洗藥物相同。因此,本課題組將常規(guī)根管沖洗藥物洗必泰與非平衡等離子體相結(jié)合,形成CHX等離子體,并通過分別應(yīng)用根管消毒封閉、非平衡等離子體和CHX等離子體三種治療措施,綜合評估其對感染根管細菌生物膜的影響。 方法:糞腸球菌或齦下菌斑懸液接種于無菌蓋玻片、牛牙本質(zhì)塊、經(jīng)過預備的單根管離體牙及牙本質(zhì)小管內(nèi),于37℃厭氧培養(yǎng),在體外建立感染根管細菌生物膜模型,分別應(yīng)用根管消毒封閉、非平衡等離子體和CHX等離子體三種治療措施進行滅菌處理。隨后,細菌活性熒光染料對處理后的細菌生物膜進行染色,立即置于激光掃描共聚焦顯微鏡下觀察,并進行三維重建,利用軟件Imaris計算得出生物膜細菌的死菌比例,從而分析這三種治療措施的滅菌效果。 結(jié)果:經(jīng)過三維重建后的感染根管細菌生物膜圖片顯示,根管消毒封閉、非平衡等離子體和CHX等離子體三種治療措施都能夠有效地殺滅絕大多數(shù)生物膜細菌;CHX等離子體可清除更多的細菌生物膜,滅菌效果明顯優(yōu)于常規(guī)根管沖洗藥物2%洗必泰和非平衡等離子體(P0.05);延長作用時間,更多的生物膜細菌被滅菌手段滅活(P0.05);相較于成熟的細菌生物膜來說,年輕的生物膜細菌對滅菌處理的抵抗性更弱,更容易被清除(P0.05)。 結(jié)論:CHX等離子體對體外的細菌生物膜具有良好的滅菌效果。在牙髓根尖周疾病治療過程中,其可以作為一種全新的輔助手段來控制細菌感染,以增加根管治療成功率,提高患牙保存率。CHX等離子體的生物相容性和對細菌生物膜的滅菌作用機理仍需進一步研究。
[Abstract]:Objective: a large number of data show that non-equilibrium plasma has a good sterilization effect. Because the gas temperature is close to room temperature, there is no obvious damage to the body tissue, the survival period of active components is short and the residue of nontoxic substances can reach small and irregular parts, its application in the field of medicine has been paid more and more attention. The early experiments of our group have proved that non-equilibrium plasma can effectively kill the biofilm of Enterococcus faecalis, and its sterilization effect is the same as that of conventional root canal washing drugs. Therefore, our group combined chlorhexidine with non-equilibrium plasma to form CHX plasma, which was treated by root canal disinfection, non-equilibrium plasma and CHX plasma, respectively.
