【摘要】：目的:研究吳茱萸堿(evodiamine,EVO)聯(lián)合射線對舌鱗癌細(xì)胞Tca-8113裸鼠移植瘤生長的抑制作用。方法:建立舌鱗癌Tca-8113細(xì)胞裸鼠皮下移植瘤模型16只,隨機(jī)分為空白對照組、EVO組、放射組及聯(lián)合組,觀察并繪制腫瘤生長曲線圖,計算腫瘤抑制率;腫瘤組織通過常規(guī)蘇木精-伊紅染色行病理學(xué)檢查;免疫組織化學(xué)法檢測移植瘤組織中Ki67、Bax和Bcl-2表達(dá)情況;透射電子顯微鏡觀察細(xì)胞凋亡形態(tài)。結(jié)果:治療第12天起,聯(lián)合組腫瘤體積與其余各組相比,明顯減小(P0.05);治療結(jié)束時,EVO組、放射組和聯(lián)合組的抑瘤率分別為15.15%、43.29%和63.20%。聯(lián)合組Ki67和Bcl-2表達(dá)水平較EVO組和放射組下降,Bax表達(dá)水平增高(P0.05)。聯(lián)合組壞死細(xì)胞數(shù)增多,且細(xì)胞呈典型凋亡特征。結(jié)論:放療聯(lián)合EVO能抑制舌鱗癌Tca-8113細(xì)胞裸鼠移植瘤的生長,可能與Ki67和Bcl-2表達(dá)下調(diào)以及Bax表達(dá)上調(diào)有關(guān)。
[Abstract]:Aim: to study the inhibitory effect of rutaecarpine (evodiamine,EVO) combined with radiation on the growth of tongue squamous cell Tca-8113 xenografts in nude mice. Methods: sixteen nude mice with tongue squamous cell carcinoma (Tca-8113) were randomly divided into three groups: blank control group, EVO group, radiation group and combined group. The tumor growth curve was observed and plotted to calculate the tumor inhibition rate. The expression of Ki67,Bax and Bcl-2 in transplanted tumor tissue was detected by immunohistochemical method and apoptosis morphology was observed by transmission electron microscope. Results: from the 12th day after treatment, the tumor volume of the combined group was significantly decreased compared with the other groups (P0.05), and at the end of the treatment, the tumor inhibition rates of the EVO group, the radiation group and the combined group were 15.15% and 63.20%, respectively. The expression of Ki67 and Bcl-2 in the combined group was lower than that in the EVO group and radiation group, and the Bax expression level was increased (P0.05). In the combined group, the number of necrotic cells increased and the cells showed typical apoptotic characteristics. Conclusion: radiotherapy combined with EVO can inhibit the growth of Tca-8113 xenografts in tongue squamous cell carcinoma, which may be related to the down-regulation of Ki67 and Bcl-2 and the up-regulation of Bax expression.
