【摘要】：目的:檢測口腔鱗狀細(xì)胞癌(oral squamous cell carcinoma,OSCC)組織中X盒結(jié)合蛋白1(X box binding protein 1,XBP1)的表達,并探討其意義。方法:應(yīng)用qRT-PCR法檢測50例OSCC腫瘤和癌旁正常黏膜上皮中剪切型XBP1(XBP1splicing,XBP1-s)的表達;應(yīng)用組織芯片技術(shù)和免疫組化染色相結(jié)合的方法檢測100例OSCC癌旁上皮、腫瘤中心、侵襲前沿及淋巴結(jié)轉(zhuǎn)移灶中XBP1蛋白的表達。結(jié)果:qRT-PCR顯示:XBP1-s mRNA表達顯著上升(log2(倍數(shù))≥1)的病例與頸淋巴結(jié)轉(zhuǎn)移相關(guān)(P0.05)。免疫組化結(jié)果顯示:XBP1蛋白在OSCC侵襲前沿和淋巴結(jié)轉(zhuǎn)移灶中的表達均高于癌旁上皮和腫瘤中心(P0.05);且在淋巴結(jié)轉(zhuǎn)移灶中的表達明顯高于侵襲前沿(P0.05)。此外,XBP1蛋白表達還與OSCC組織病理學(xué)分級相關(guān)(P0.05)。結(jié)論:XBP1可能是促進口腔鱗狀細(xì)胞癌侵襲和轉(zhuǎn)移的一個重要因子。
[Abstract]:Aim: to investigate the expression of X-cassette binding protein 1 (X box binding protein 1 (XBP1) in oral squamous cell carcinoma (oral squamous cell carcinoma,OSCC) and its significance. Methods: qRT-PCR method was used to detect the expression of XBP1 (XBP1splicing,XBP1-s) in 50 cases of OSCC tumor and adjacent normal mucosal epithelium.
