下調(diào)α-catulin基因的表達(dá)對舌鱗癌細(xì)胞株Tscca侵襲及遷移能力影響的體外研究

【摘要】:目的探討α-catulin基因下調(diào)對人舌鱗癌細(xì)胞株Tscca侵襲、遷移能力的影響。方法本實(shí)驗(yàn)設(shè)為3個(gè)組:α-catulin-siRNA組、空質(zhì)粒轉(zhuǎn)染組和未轉(zhuǎn)染質(zhì)粒的空白對照組。構(gòu)建α-catulin沉默質(zhì)粒轉(zhuǎn)染的人舌鱗癌細(xì)胞株Tscca,運(yùn)用Western blotting檢測α-catulin蛋白表達(dá),采用Transwell及劃痕實(shí)驗(yàn)檢測細(xì)胞的侵襲、遷移能力。結(jié)果 Western blotting結(jié)果顯示,α-catulin-siRNA轉(zhuǎn)染組Tscca細(xì)胞α-catulin蛋白表達(dá)量減少;Transwell及劃痕實(shí)驗(yàn)結(jié)果顯示,α-catulin-siRNA轉(zhuǎn)染組細(xì)胞侵襲、遷移能力降低。結(jié)論下調(diào)α-catulin基因的表達(dá)可以有效降低人舌鱗癌細(xì)胞株Tscca的侵襲、遷移能力,α-catulin有望成為舌癌基因治療的一個(gè)潛在靶點(diǎn)。
[Abstract]:Objective to investigate the effect of down-regulation of 偽-catulin gene on the invasion and migration of human tongue squamous cell carcinoma cell line Tscca. Methods the experiment was divided into three groups: 偽-catulin-siRNA group, empty plasmid transfection group and blank control group. Human tongue squamous cell carcinoma cell line Tscca, transfected with 偽-catulin silencing plasmid was constructed to detect the expression of 偽-catulin protein by Western blotting, and the invasion and migration ability of the cells were detected by Transwell and scratch test. Results the results of Western blotting showed that the expression of 偽-catulin protein decreased in Tscca cells transfected with 偽-catulin-siRNA, and the expression of 偽-catulin protein in Tscca cells was decreased by Transwell and scratch test. The results of Transwell and scratch test showed that the cell invasion and migration ability of 偽-catulin-siRNA transfected group were decreased. Conclusion down-regulation of 偽-catulin gene expression can effectively reduce the invasion and migration ability of human tongue squamous cell carcinoma cell line Tscca. 偽-catulin may be a potential target of tongue oncogene therapy.
