正畸移動(dòng)大鼠牙過(guò)程牙周組織中PLAP-1、BMP-2的表達(dá)及意義

【摘要】:目的:研究正畸移動(dòng)大鼠上后牙過(guò)程中,PLAP-1、BMP-2在牙周膜的分布和表達(dá)。方法:選擇雄性SD大鼠36只,按1d、3d、5d、7d、14d、21d分為6組,均用50g移動(dòng)左上第一磨牙,右上第一磨牙為對(duì)照。在相應(yīng)時(shí)期處死大鼠后測(cè)量牙移動(dòng)距離,采用HE染色及免疫組織化學(xué)SP法,檢測(cè)不同時(shí)期牙周組織的形態(tài)變化;PLAP-1、BMP-2在牙周膜組織中的表達(dá)及研究?jī)烧呖赡艽嬖诘年P(guān)系。結(jié)果:不同時(shí)期牙移動(dòng)的距離不同,差別顯著有統(tǒng)計(jì)學(xué)意義(P0.05)。HE染色顯示對(duì)照組牙周組織變化不明顯;實(shí)驗(yàn)組張力區(qū)有成骨細(xì)胞生成,壓力區(qū)破骨細(xì)胞生成,隨著時(shí)間的延長(zhǎng)最后兩區(qū)形態(tài)都基本趨一致。免疫組化法示PLAP-1在牙周膜的表達(dá)總是張力區(qū)高于壓力區(qū),在第3天張力區(qū)表達(dá)最強(qiáng),除21d外,其余各組間差異顯著有統(tǒng)計(jì)學(xué)意義(P0.05)。BMP-2在第5天張力側(cè)表達(dá)最強(qiáng),在3d、5d、7d的表達(dá)差異顯著有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:PLAP-1,BMP-2在正畸力作用下參與牙周膜組織改建的表達(dá)最高值時(shí)期不同。
[Abstract]:Objective: to study the distribution and expression of PLAP-1,BMP-2 in periodontal ligament during orthodontic movement of rat upper posterior teeth. Methods: Thirty-six male SD rats were divided into 6 groups according to 1 d, 3 d, 5 d, 7 d, 14 d and 21 d, all of them were divided into 6 groups. The left first molar was moved 50 g and the right first molar was used as control. After the rats were killed at the corresponding period, the distance of tooth movement was measured, the morphological changes of periodontal tissue were detected by HE staining and immunohistochemical SP method, and the expression of PLAP-1,BMP-2 in periodontal ligament tissue and the possible relationship between them were studied. Results: the distance of tooth movement in different period was different, the difference was statistically significant (P0.05). HE staining showed that the periodontal tissue of the control group had no obvious change, the tension zone of experimental group had osteoblast formation, the pressure zone of osteoclast formation, and the change of osteoclast formation in the pressure area. With the extension of time, the last two regions are basically consistent. Immunohistochemical method showed that the expression of PLAP-1 in periodontal ligament was always higher than that in the pressure area, and the expression of BMP-2 was the strongest in the tension area on the 3rd day, except 21 days, there was significant difference between the other groups (P0.05). The expression of BMP-2 was the strongest in the tension side of the periodontal ligament on the 5th day. There was significant difference in the expression of 7 days after 3 days (P 0.05). Conclusion: the maximum expression of PLAP-1,BMP-2 in periodontal ligament tissue remodeling under orthodontic force is different.
