高齲學(xué)齡前兒童變形鏈球菌的水平傳播方式

【摘要】:目的研究高齲學(xué)齡前兒童變形鏈球菌的水平傳播方式,為兒童齲病早期預(yù)防提供依據(jù)。方法收集20名4~5歲高齲學(xué)齡前兒童口腔中牙菌斑標本進行厭氧培養(yǎng),通過細菌形態(tài)學(xué)觀察、生物化學(xué)鑒定、聚合酶鏈反應(yīng)(PCR)擴增變形鏈球菌基因的特異性片段葡糖基轉(zhuǎn)移酶B(gtfB)以及隨機引物聚合酶鏈反應(yīng)(AP-PCR)等方法對獲得的變形鏈球菌臨床分離株進行鑒定。結(jié)果從20例高齲學(xué)齡前兒童口腔中分離獲得200株變形鏈球菌臨床分離株,經(jīng)生物化學(xué)鑒定排除其中19株,獲得181株變形鏈球菌臨床分離株,變形鏈球菌的特異性基因gtfB的PCR擴增181株變形鏈球菌臨床分離株均鑒定為變形鏈球菌,AP-PCR發(fā)現(xiàn)37種不同基因型,其中6名兒童攜帶1種基因型,11名兒童攜帶2種基因型,2名兒童攜帶3種基因型,1名兒童攜帶4種基因型,有2名不同班級兒童攜帶一種相同基因型。結(jié)論獲得37株不同基因型變形鏈球菌臨床分離株,高齲學(xué)齡前兒童間未發(fā)現(xiàn)變形鏈球菌水平傳播。
[Abstract]:Objective to study the horizontal transmission of Streptococcus mutans in preschool children and to provide evidence for early prevention of caries in children. Methods 20 dental plaque specimens from the oral cavity of 20 children aged 4 to 5 years old with high caries were collected for anaerobic culture. The bacterial morphology was observed and identified by biochemistry. Polymerase chain reaction (PCR) was used to amplify the specific fragment of Streptococcus mutans gene glucosyltransferase (B (gtfB) and random primer polymerase chain reaction (AP-PCR) to identify the clinical isolates of Streptococcus mutans. Results 200 strains of Streptococcus mutans were isolated from the oral cavity of 20 children with high caries. Among them, 19 strains were excluded by biochemistry, and 181 strains of Streptococcus mutans were obtained. The clinical isolates of Streptococcus mutans were identified by PCR amplification of the specific gene gtfB of Streptococcus mutans and 37 different genotypes were identified by AP-PCR. Among them, 6 children carry one genotype, 11 children carry 2 genotypes, 2 children carry 3 genotypes, 1 child carries 4 genotypes, and 2 children in different classes carry one of the same genotypes. Conclusion among 37 clinical isolates of Streptococcus mutans with different genotypes, no horizontal transmission of Streptococcus mutans was found among preschool children with high caries.
