黃芩苷對(duì)糞腸球菌毒力因子作用的體外研究

【摘要】:目的:研究黃芩苷對(duì)體外糞腸球菌生物膜的抑菌效果,并探討黃芩苷對(duì)糞腸球菌毒力因子asal、EfaA、ebpA基因轉(zhuǎn)錄表達(dá)水平的影響。方法:6孔板建立糞腸球菌體外生物膜模型,利用激光共聚焦顯微鏡觀察不同濃度黃芩苷作用24 h后的抑菌效果;然后再根據(jù)抑菌效果,選擇適宜濃度(2、4、8 mg/mL)的黃芩苷分別與糞腸球菌共同培養(yǎng)12 h,并利用RT-PCR技術(shù)檢測(cè)各組糞腸球菌毒力因子asal、EfaA、ebpA的表達(dá)水平。結(jié)果:激光共聚焦顯微鏡觀察顯示:黃芩苷對(duì)糞腸球菌生物膜的抑菌作用隨藥物濃度的增加而增加,8 mg/mL時(shí),生物膜內(nèi)的活菌比例下降最明顯;RT-PCR檢測(cè)顯示:與陰性對(duì)照組相比,2、4、8 mg/mL的黃芩苷均能明顯抑制asal、EfaA、ebpA基因的表達(dá)(P0.05),其抑制作用隨濃度增加而增加,各濃度組兩兩相比均有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論:黃芩苷對(duì)糞腸球菌生物膜及其毒力因子asal、EfaA、ebpA均有明顯的抑制作用。
[Abstract]:Aim: to study the bacteriostatic effect of baicalin on Enterococcus faecalis biofilm in vitro and to investigate the effect of baicalin on the transcription level of virulence factor asal,EfaA,ebpA gene of Enterococcus faecalis. Methods the in vitro biofilm model of Enterococcus faecalis was established with the 6 pore plate. The bacteriostatic effect of baicalin at different concentrations was observed by confocal laser microscope after 24 h, and then according to the bacteriostatic effect. Baicalin was cultured with Enterococcus faecalis for 12 hours, and the expression of virulence factor asal,EfaA,ebpA in each group was detected by RT-PCR technique. Results: the results of laser confocal microscopy showed that the bacteriostatic effect of baicalin on the biofilm of Enterococcus faecalis increased with the increase of drug concentration for 8 mg/mL. The results of RT-PCR showed that baicalin at 8 mg/mL could significantly inhibit the expression of asal,EfaA,ebpA gene (P0.05). The inhibitory effect of baicalin increased with the increase of concentration, and there was significant difference between the two groups (P0.05). Conclusion: baicalin has obvious inhibitory effect on Enterococcus faecalis biofilm and virulence factor asal,EfaA,ebpA.
