【摘要】：目的:探討高糖環(huán)境下不同濃度BMP-7對成骨細胞生物學性能的影響。方法:將成骨細胞(MC3T3-E1Subclone 14)在葡萄糖(25 mmol/L)環(huán)境下培養(yǎng),據(jù)BMP-7濃度分為0(對照組)、50、100、200 ng/ml(實驗組)4組。檢測細胞增殖及ALP活性、礦化結節(jié)、成骨基因Runx2及OCN mRNA的表達;培養(yǎng)24 h觀察細胞骨架的形態(tài)變化。結果:BMP-7可顯著促進細胞增殖及ALP活性(P0.05)。礦化結節(jié)數(shù)量增多、體積增大明顯。實驗組Runx2、OCN mRNA水平明顯上調(P0.05);200 ng/ml BMP-7組Runx2、OCN mRNA的水平較其他組同一時間點顯著上調(P0.05)。細胞骨架漸呈網格狀鋪展且隨BMP-7濃度增高而明顯。結論:BMP-7可抑制高糖對MC3T3-E1的作用,提示BMP-7有利于促進糖尿病患者種植體的骨結合。
[Abstract]:Objective: to investigate the effects of different concentrations of BMP-7 on the biological properties of osteoblasts. Methods: osteoblasts (MC3T3-E1Subclone 14) were cultured in glucose (25 mmol/L). According to the concentration of BMP-7, osteoblasts were divided into 4 groups: 0 (control group) 50100200 ng/ml (experimental group). Cell proliferation and ALP activity, mineralized nodules, expression of osteogenic gene Runx2 and OCN mRNA were detected, and morphological changes of cytoskeleton were observed after 24 h culture. Results: the cell proliferation and ALP activity were significantly increased by 1% BMP-7 (P 0.05). The number and volume of mineralized nodules increased obviously. The level of Runx2,OCN mRNA was significantly up-regulated in the experimental group (P0.05). The level of Runx2,OCN mRNA in the 200 ng/ml BMP-7 group was significantly higher than that in the other groups at the same time point (P0.05). The cytoskeleton gradually spread out in the shape of grid and was obvious with the increase of BMP-7 concentration. ConclusionBMP-7 can inhibit the effect of high glucose on MC3T3-E1, suggesting that BMP-7 can promote bone bonding of implants in diabetic patients.
