【摘要】：目的：獲得高轉移舌鱗狀細胞癌細胞,為建立高轉移舌癌動物模型奠定基礎。 方法：采用改良侵襲實驗篩選出高低不同的轉移潛能細胞；采用MTT實驗和平板克隆實驗檢測兩種細胞的克隆差異；侵襲性實驗和遷移性實驗檢測兩種細胞侵襲和轉移差異；流式細胞儀用來檢測兩種細胞生長周期的差異；采用Western Blot和Real-time PCR方法檢測兩種細胞Zeste基因增強子同源物2(EZH2)的蛋白和mRNA表達差異。 結果：分離出兩種不同轉移潛能細胞。兩種不同轉移細胞的克隆形成、侵襲和遷移、細胞周期和EZH2蛋白和mRNA表達均有顯著性差異(t=3.423～59.300,P0.05～0.001)。 結論：從舌鱗癌細胞系中分離得到了兩種不同轉移潛能的細胞亞群,且兩種細胞具有不同的生物學特性。
[Abstract]:Objective: to obtain high metastatic squamous cell carcinoma of tongue and to establish animal model of high metastasis of tongue cancer. Methods: different metastatic potential cells were screened by modified invasion assay, clone differences were detected by MTT assay and plate cloning assay, invasion and metastasis were detected by invasive assay and migration assay. Flow cytometry was used to detect the difference of cell growth cycle, and Western Blot and Real-time PCR methods were used to detect the protein and mRNA expression of Zeste gene enhancer homologue 2 (EZH2). Results: two different metastatic potential cells were isolated. There were significant differences in cell cycle, EZH2 protein and mRNA expression between the two kinds of metastatic cells. Conclusion: two kinds of cell subsets with different metastatic potential were isolated from tongue squamous cell carcinoma cell lines, and the two kinds of cells had different biological characteristics.
