【摘要】：目的：獲得高轉(zhuǎn)移舌鱗狀細(xì)胞癌細(xì)胞,為建立高轉(zhuǎn)移舌癌動(dòng)物模型奠定基礎(chǔ)。 方法：采用改良侵襲實(shí)驗(yàn)篩選出高低不同的轉(zhuǎn)移潛能細(xì)胞；采用MTT實(shí)驗(yàn)和平板克隆實(shí)驗(yàn)檢測(cè)兩種細(xì)胞的克隆差異；侵襲性實(shí)驗(yàn)和遷移性實(shí)驗(yàn)檢測(cè)兩種細(xì)胞侵襲和轉(zhuǎn)移差異；流式細(xì)胞儀用來(lái)檢測(cè)兩種細(xì)胞生長(zhǎng)周期的差異；采用Western Blot和Real-time PCR方法檢測(cè)兩種細(xì)胞Zeste基因增強(qiáng)子同源物2(EZH2)的蛋白和mRNA表達(dá)差異。 結(jié)果：分離出兩種不同轉(zhuǎn)移潛能細(xì)胞。兩種不同轉(zhuǎn)移細(xì)胞的克隆形成、侵襲和遷移、細(xì)胞周期和EZH2蛋白和mRNA表達(dá)均有顯著性差異(t=3.423～59.300,P0.05～0.001)。 結(jié)論：從舌鱗癌細(xì)胞系中分離得到了兩種不同轉(zhuǎn)移潛能的細(xì)胞亞群,且兩種細(xì)胞具有不同的生物學(xué)特性。
[Abstract]:Objective: to obtain high metastatic squamous cell carcinoma of tongue and to establish animal model of high metastasis of tongue cancer. Methods: different metastatic potential cells were screened by modified invasion assay, clone differences were detected by MTT assay and plate cloning assay, invasion and metastasis were detected by invasive assay and migration assay. Flow cytometry was used to detect the difference of cell growth cycle, and Western Blot and Real-time PCR methods were used to detect the protein and mRNA expression of Zeste gene enhancer homologue 2 (EZH2). Results: two different metastatic potential cells were isolated. There were significant differences in cell cycle, EZH2 protein and mRNA expression between the two kinds of metastatic cells. Conclusion: two kinds of cell subsets with different metastatic potential were isolated from tongue squamous cell carcinoma cell lines, and the two kinds of cells had different biological characteristics.
