【摘要】：目的:探討串珠素在牙髓損傷修復(fù)過程中的表達(dá)變化及其可能的作用。方法:取雄性SD大鼠48只,隨機(jī)分為4組(n=12)。建立大鼠一側(cè)上頜第一磨牙牙髓損傷模型,另一側(cè)同名牙作為正常對照組;分別于損傷后1、3、5、7 d各取1組目標(biāo)牙樣本,采用免疫組化染色和Real-time PCR觀察檢測各組牙髓組織中串珠素的表達(dá)變化,并采用單因素方差分析比較各組間的差異。結(jié)果:免疫組化染色顯示,牙髓損傷后,牙髓組織中串珠素染色強(qiáng)度增加,各實(shí)驗組的平均光密度均明顯高于正常對照組(P0.05),并以1 d組升高最為明顯;RT-PCR檢測顯示,與正常對照組相比,串珠素mRNA的相對表達(dá)量在牙髓損傷后1 d明顯下降,此后隨時間延長逐漸回升,損傷后7 d時回升最明顯,但仍明顯低于對照組(P0.05)。結(jié)論:串珠素在牙髓組織損傷后的防御機(jī)制中起一定的作用,但其表達(dá)的增加并不是由其生物合成增加所致。
[Abstract]:Objective: to investigate the expression and possible role of series beads in the process of pulp injury and repair. Methods: 48 male SD rats were randomly divided into 4 groups (n = 12). The pulp injury model of one maxillary first molar was established in rats, and the other side of the same name was used as the normal control group. Immunohistochemical staining and Real-time PCR were used to observe the changes of the expression of series globin in the pulp tissue of each group, and the differences between the groups were compared by univariate analysis of variance (ANOVA). Results: immunohistochemical staining showed that the intensity of string bead staining in pulp tissue increased after pulp injury, and the average optical density of each experimental group was significantly higher than that of normal control group (P0.05). Compared with the normal control group, the relative expression of mRNA decreased significantly at 1 day after pulp injury, and then gradually increased with time, and increased significantly at 7 days after injury, but still significantly lower than that in the control group (P0.05). Conclusion: series beads may play an important role in the defense mechanism of pulp injury, but the increase of its expression is not caused by the increase of biosynthesis.
