氟環(huán)境下變形鏈球菌耐氟菌株ciaH、eno和pykF基因的差異表達(dá)及其意義

【摘要】:目的:檢測變形鏈球菌耐氟菌株在氟環(huán)境下ciaH、eno和pykF基因表達(dá)的差異,探討ciaH、eno和pykF基因與變形鏈球菌氟抗性的關(guān)系。方法:變形鏈球菌親代菌株及其耐氟菌株,分為親代組(變形鏈球菌親代菌株生長于無氟BHI培養(yǎng)基中)、耐氟組(耐氟菌株生長于無氟BHI培養(yǎng)基中)和含氟組(耐氟菌株生長于含氟量為1g·L~(-1)的BHI培養(yǎng)基中)。分別選取3組處于生長對數(shù)期(11h)和穩(wěn)定期(20h)的菌株,采用RT-PCR法檢測ciaH、eno和pykF mRNA表達(dá)水平。結(jié)果:與耐氟組比較,含氟組的耐氟菌株ciaH、eno和pykF mRNA表達(dá)水平在對數(shù)期和穩(wěn)定期均明顯升高(P0.05或P0.01);與親代組比較,耐氟組的耐氟菌株eno和pykF mRNA表達(dá)水平在對數(shù)期和穩(wěn)定期均明顯降低(P0.01),ciaH mRNA表達(dá)水平在對數(shù)期差異無統(tǒng)計(jì)學(xué)意義(P0.05),在穩(wěn)定期明顯升高(P0.01)。結(jié)論:氟能提高耐氟菌株ciaH、eno和pykF基因的表達(dá),該組基因與耐氟菌株氟抗性的產(chǎn)生有關(guān)聯(lián)。
[Abstract]:Objective: to detect the difference of the expression of Cia Hneno and pykF genes between Streptococcus mutans and Streptococcus mutans in fluorine environment, and to explore the relationship between Cia Hneno and pykF genes and the fluorine resistance of Streptococcus mutans. Methods: Streptococcus mutans parents and fluorine tolerant strains were used. It was divided into three groups: parent group (Streptococcus mutans grew in BHI medium without fluorine), fluorine tolerant group (fluorine tolerant strain grew in BHI medium without fluoride) and fluorine group (fluoride tolerant strain grew in BHI medium with fluorine content of 1 g L ~ (-1). Three groups of strains in logarithmic phase (11 h) and stable stage (20 h) were selected to detect the expression of Cia Hmeno and pykF mRNA by RT-PCR method. Results: compared with the fluorine-tolerant group, the expression levels of ciaHeno and pykF mRNA in fluorine-tolerant strain were significantly increased in logarithmic phase and stable period (P0.05 or P0.01), and compared with parent group, the expression level of Cia Hneno and pykF mRNA in fluorine-tolerant group were significantly higher than those in control group (P0.05 or P0.01). The expression levels of eno and pykF mRNA in fluorine-tolerant strains were significantly decreased in logarithmic phase and stable phase (P0.01). There was no significant difference in the expression level of ciaH mRNA in logarithmic phase (P0.05), but significantly increased in stable phase (P0.01). Conclusion: fluorine can enhance the expression of Cia Hneno and pykF genes in fluorine tolerant strains, which is related to the production of fluorine resistance in fluorine tolerant strains.
