牙釉質(zhì)結(jié)合肽的體外礦化研究

【摘要】:本研究首次報道具有牙釉質(zhì)結(jié)合活性的環(huán)肽序列(-CMPQVMPMC-)。在反篩試驗中,投入等量牙釉質(zhì)粉末和相同滴度噬菌體,外殼蛋白pⅢ上表達(dá)該展示肽的噬菌體回收量明顯高于不含展示肽的野生型噬菌體M13。固相法體外合成該環(huán)狀七肽以模擬展示肽在噬菌體表面的構(gòu)象,并利用96孔板體外快速凝膠單擴(kuò)散系統(tǒng)驗證其對鈣/磷沉積的影響。礦化開始后的24h內(nèi),每隔6h記錄各樣本在405nm的OD值。各樣本的相對濁度表示為所占24h末空白組OD值增量(100%)的百分比。結(jié)果顯示,牙釉質(zhì)結(jié)合肽(EBP)能明顯延遲凝膠中的鈣磷成核,且延遲效果與添加的濃度相關(guān)。在礦化24h末,含結(jié)合肽的各組凝膠內(nèi)濁度明顯低于空白組,但高于BSA組。
