生理性根吸收過程中乳牙牙髓干細(xì)胞通過α7 nAChR調(diào)節(jié)破骨細(xì)胞分化能力的研究

【摘要】:目的:探討生理性根吸收過程中乳牙牙髓干細(xì)胞(DDPSCs)通過α7亞型煙堿型乙酰膽堿受體(α7 nAChR)對破骨分化能力的調(diào)節(jié)作用。方法:酶消化法和有限稀釋法分離培養(yǎng)根吸收不同時期DDPSCs和恒牙牙髓干細(xì)胞(DPSCs)。采用實時定量PCR和Western blot,檢測各組細(xì)胞α7 nAChR、RANKL、OPG的表達及其比值差異,以及阻斷α7 nAChR后RANKL、OPG的表達及其比值變化。結(jié)果:α7 nAChR表達及RANKL/OPG比值在根吸收中期DDPSCs中明顯升高(P0.05)。阻斷α7nAChR后,RANKL/OPG明顯下降(P0.05)。結(jié)論:在乳牙生理性根吸收過程中,α7nAChR通過上調(diào)自身表達,參與對RANKL/OPG的調(diào)節(jié),影響DDPSCs的破骨分化能力。
[Abstract]:Aim: to investigate the effect of primary dental pulp stem cells (DDPSCs) on osteoclast differentiation through 偽 7 subtype nicotinic acetylcholine receptor (偽 7 nAChR) during physiological root resorption. Methods: DDPSCs and DPSCs were isolated and cultured by enzyme digestion and limited dilution. Real time quantitative PCR and Western blot were used to detect the expression and ratio difference of 偽 7 nAChR RANKL OPG, and the expression and ratio of RANKL1 OPG after blocking 偽 7 nAChR. Results: the expression of 偽 7 nAChR and the ratio of RANKL / OPG were significantly increased in DDPSCs (P0.05). After blocking 偽 7 nAChR, RANKL / OPG decreased significantly (P 0.05). Conclusion: during the process of physiologic root resorption of deciduous teeth, 偽 7nAChR participates in the regulation of RANKL / OPG by upregulating its own expression, thus affecting the osteoclastic differentiation of DDPSCs.
