【摘要】：目的:研究萊菔硫烷(SFN)對涎腺腺樣囊性ACC-M癌細(xì)胞增殖和凋亡的影響。方法:分別用終濃度為5、10、20、30、40、60、80、100μmol/L的SFN處理ACC-M細(xì)胞24、48、72 h。用MTT比色試驗和臺盼藍(lán)拒染實驗檢測細(xì)胞的增殖和存活情況;倒置顯微鏡、Giemsa染色、透射電鏡觀察細(xì)胞形態(tài)學(xué)變化;Annexin-V-FITC和PI雙標(biāo)記流式細(xì)胞儀檢測細(xì)胞凋亡情況。結(jié)果:SFN對ACC-M有明顯的增殖抑制作用,最高抑制率達(dá)89.2%,作用24、48、72 h時的IC50(μmol/L)分別是75.6、21.3、16.5,增殖抑制率與藥物濃度和作用時間具有明顯的正相關(guān)性。SFN可誘導(dǎo)ACC-M細(xì)胞凋亡,凋亡率隨處理時間延長和藥物濃度增加而上升(P0.01)。結(jié)論:SFN具有抑制涎腺腺樣囊性癌細(xì)胞系A(chǔ)CC-M增殖和誘導(dǎo)其凋亡的作用,且具有濃度和時間依賴性。
[Abstract]:Aim: to study the effects of sulforaphane (SFN) on the proliferation and apoptosis of salivary adenoid cystic ACC-M cells. Methods: ACC-M cells were treated with SFN at the final concentration of 5 渭 mol / L, 10, 10, 20, 30, 40, 60, 100 渭 mol / L, for 24, 48 and 72 hours, respectively. MTT colorimetric assay and trypan blue exclusion assay were used to detect cell proliferation and survival, and cell apoptosis was detected by inverted microscope Giemsa staining, transmission electron microscopy and Annexin-V-FITC and Pi double labeling flow cytometry. Results the cell proliferation of ACC-M was inhibited by 1% SFN with the highest inhibitory rate of 89.2.The IC50 (渭 mol / L) was 75.6 渭 mol / L for 72 h, respectively. The inhibitory rate of proliferation was positively correlated with the concentration and the time of action. SFN could induce apoptosis of ACC-M cells, and the IC50 (渭 mol / L) was 75.6 渭 mol / L for 72 h. SFN could induce the apoptosis of ACC-M cells. The apoptotic rate increased with the prolongation of treatment time and the increase of drug concentration (P0.01). Conclusion% SFN can inhibit the proliferation and induce apoptosis of salivary adenoid cystic carcinoma cell line ACC-M in a concentration and time dependent manner.
