【摘要】：目的:體外培養(yǎng)鼠根尖乳頭干細(xì)胞和牙周膜干細(xì)胞,比較研究腫瘤壞死因子α對兩者增殖活性的影響,探討年輕恒牙在炎癥條件下,兩者用于牙根再生的可能性。方法:取Wistar幼鼠根尖尚未發(fā)育完全的健康下頜牙,酶消化法獲得根尖乳頭干細(xì)胞和牙周膜干細(xì)胞,免疫熒光法鑒定兩者干細(xì)胞表面標(biāo)志物STRO-1的表達(dá)。選取第2代根尖乳頭干細(xì)胞和牙周膜干細(xì)胞的單細(xì)胞懸液,采用MTT法比較兩者的生長曲線,繪制生長曲線圖。將含有腫瘤壞死因子α濃度為0、5、10、50ng/m L的培養(yǎng)基加入根尖乳頭干細(xì)胞和牙周膜干細(xì)胞的第3代細(xì)胞中,使用MTT法檢測兩者各孔吸光密度值,比較其對兩種細(xì)胞增殖活性的影響。結(jié)果:1.根尖乳頭干細(xì)胞和牙周膜干細(xì)胞均呈典型的成纖維細(xì)胞樣的梭形結(jié)構(gòu)。2.根尖乳頭干細(xì)胞和牙周膜干細(xì)胞均表達(dá)干細(xì)胞特性;3.根尖乳頭干細(xì)胞和牙周膜干細(xì)胞各光孔密度值隨時間的遞增而呈現(xiàn)出增高的趨勢。自第4天起根尖乳頭干細(xì)胞的增殖活力明顯強于牙周膜干細(xì)胞。4.在10ng/mL和50ng/mL濃度時,腫瘤壞死因子α能明顯促進(jìn)根尖乳頭干細(xì)胞的增殖,亦能明顯促進(jìn)牙周膜干細(xì)胞的增殖。在10ng/mL和50ng/mL濃度時,根尖乳頭干細(xì)胞的增殖能力強于牙周膜干細(xì)胞。結(jié)論:根尖乳頭干細(xì)胞在正常狀態(tài)和炎癥狀態(tài)下增殖活性均強于牙周膜干細(xì)胞,此研究結(jié)果為種子細(xì)胞在年輕恒牙發(fā)生根尖周炎癥后牙根繼續(xù)發(fā)育的能力提供實驗依據(jù)。
[Abstract]:Objective: to culture rat apical papilla stem cells and periodontal ligament stem cells in vitro, to compare the effects of tumor necrosis factor 偽 on the proliferation of the two cells, and to explore the possibility of the young permanent teeth being used for root regeneration under the condition of inflammation. Methods: the apical papillary stem cells and periodontal ligament stem cells were obtained from immature healthy mandibular teeth of young Wistar rats. The expression of STRO-1 on the surface of stem cells was identified by immunofluorescence. Single cell suspensions of apical papilla stem cells and periodontal ligament stem cells of the second generation were selected and the growth curves of the two cells were compared by MTT method and the growth curves were plotted. The culture medium containing 50 ng / mL TNF- 偽 was added to the third passage cells of apical papillary stem cells and periodontal ligament stem cells. MTT assay was used to detect the absorptivity of the two cells and to compare their effects on the proliferative activity of the two kinds of cells. The result is 1: 1. The apical papilla stem cells and periodontal ligament stem cells were both typical fibroblast-like fusiform structures. Apical papilla stem cells and periodontal ligament stem cells both expressed stem cell characteristics. The optical pore density of apical papilla stem cells and periodontal ligament stem cells increased with time. The proliferative activity of apical papilla stem cells was significantly stronger than that of periodontal ligament stem cells. At the concentrations of 10 ng / mL and 50 ng / mL, TNF- 偽 significantly promoted the proliferation of apical papilla stem cells and periodontal ligament stem cells. At the concentrations of 10 ng / mL and 50 ng / mL, the proliferation of apical papillary stem cells was stronger than that of periodontal ligament stem cells. Conclusion: the proliferative activity of apical papillary stem cells is stronger than that of periodontal ligament stem cells in normal and inflammatory state. The results provide experimental evidence for the ability of seed cells to continue to develop after periapical inflammation in young permanent teeth.
