基質(zhì)膠緩釋神經(jīng)軸突導(dǎo)向分子SEMA3A對(duì)去勢(shì)大鼠鈦種植體穩(wěn)定性的影響

【摘要】:目的:評(píng)估基質(zhì)膠緩釋SEMA3A對(duì)去勢(shì)大鼠種植體穩(wěn)定性的影響。方法:對(duì)60只SD大鼠行雙側(cè)卵巢切除術(shù)。12周后,根據(jù)植入種植體的不同將動(dòng)物隨機(jī)分為3組:1)對(duì)照組,種植體+蒸餾水;2)基質(zhì)膠組,種植體+基質(zhì)膠;3)種植體+基質(zhì)膠+SEMA3A,將種植體植入所有動(dòng)物脛骨近端的干骺端。在利用酶聯(lián)免疫吸附試驗(yàn)檢測(cè)種植體表面基質(zhì)膠緩釋SEMA3A的生物學(xué)過程進(jìn)行檢測(cè)。種植體植入后的第4周和第8周將脛骨與植入體一起收集,行顯微CT、組織學(xué)和生物力學(xué)檢測(cè)。結(jié)果:SEMA3A的體外釋放檢測(cè)發(fā)現(xiàn),連續(xù)10d可以檢測(cè)到基質(zhì)膠中釋放的SEMA3A,其中前3d呈爆發(fā)性釋放過程;種植體植入后第4和第8周,與單獨(dú)使用基質(zhì)膠組相比,SEMA3A+基質(zhì)膠組骨體積百分比分別增加了88.7%和83.3%,骨小梁粗度分別增加了51.4%和49.9%,骨小梁數(shù)量分別增加了54.2%和62.1%;在組織學(xué)定量評(píng)價(jià)中,種植體周圍相對(duì)骨面積分別增加了89.8%和149.2%,骨與種植體結(jié)合率分別增加了148.9%和24.8%;生物力學(xué)檢測(cè)發(fā)現(xiàn),種植體的最大推出力增加了149.3%和209.2%,結(jié)合剛度增加171.2%和210.6%,結(jié)合韌性增加了151.8%和268.2%。結(jié)論:用SEMA3A加基質(zhì)膠對(duì)種植體進(jìn)行表面改性可以有效改善去勢(shì)大鼠種植體的骨整合與穩(wěn)定性。
[Abstract]:Objective: To evaluate the effect of matrix gel sustained release SEMA3A on the stability of implants in ovariectomized rats. Methods: after.12 weeks of bilateral oophorectomy in 60 SD rats, the animals were randomly divided into 3 groups according to the implantation implants: 1) the control group, the implant + distilled water; 2) matrix gel, implant + matrix glue; 3) implant + matrix glue +SEMA3A, which would implant the implant. The metaphysis of the proximal tibia of all animals was implanted. The biological process of detecting the sustained-release SEMA3A on the surface of the implant surface was detected by enzyme linked immunosorbent assay. The tibia was collected with the implant at fourth and eighth weeks after implantation. The microscopical CT, histology and Biodynamics test were performed. Results: the release of SEMA3A in vitro was detected. It was found that continuous 10d could detect the release of SEMA3A in matrix glue, in which the former 3D showed an explosive release process, and fourth and eighth weeks after implantation, the percentage of bone volume increased by 88.7% and 83.3% in the SEMA3A+ matrix group, respectively, and the bone trabecular size increased by 51.4% and 49.9% respectively, and the number of trabeculae increased respectively. In the quantitative evaluation of histology, the relative bone area around the implant was increased by 89.8% and 149.2%, and the bone and implant binding rate increased by 148.9% and 24.8%, respectively. The maximum release strength of the implant was increased by 149.3% and 209.2%, the stiffness increased by 171.2% and 210.6%, and the toughness was increased by 151.8% in combination with the toughness. And 268.2%. conclusion: the surface modification of the implant with SEMA3A plus matrix adhesive can effectively improve the osseointegration and stability of the implant in ovariectomized rats.
