【摘要】：目的從基因和蛋白水平研究增殖細(xì)胞核抗原(PCNA)和P53在萎縮性腮腺再生過(guò)程中的表達(dá)及意義。方法將102只Wistar大鼠分為實(shí)驗(yàn)組和對(duì)照組,結(jié)扎實(shí)驗(yàn)組大鼠腮腺主導(dǎo)管,分別于結(jié)扎7 d(A組)、14 d(B組)后實(shí)現(xiàn)腮腺導(dǎo)管再通,并于再通后第0、3、5、7、10、14、21、28天處死大鼠獲取新鮮腮腺組織標(biāo)本。采用蘇木精-伊紅(HE)染色法觀察腮腺組織形態(tài)學(xué)變化,實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)和Western blot法檢測(cè)兩組腮腺組織中PCNA和P53的表達(dá)。結(jié)果兩實(shí)驗(yàn)組在腮腺導(dǎo)管結(jié)扎第7、14天,腺泡凋亡,導(dǎo)管細(xì)胞增殖,同組P53相對(duì)表達(dá)量高于PCNA;隨著再通時(shí)間延長(zhǎng),腺泡細(xì)胞逐漸增多,A組在導(dǎo)管再通后第3天、B組在導(dǎo)管再通后第5天,PCNA和P53相對(duì)表達(dá)量達(dá)到峰值,與對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.01);其后PCNA和P53相對(duì)表達(dá)量逐漸減少,A組于再通后第21天、B組于再通后第28天接近對(duì)照組。結(jié)論腮腺導(dǎo)管結(jié)扎后,P53相對(duì)表達(dá)量增多,誘導(dǎo)腮腺腺泡凋亡;導(dǎo)管再通后,PCNA相對(duì)表達(dá)量逐漸增多,達(dá)峰值后減少,可促進(jìn)腺泡增殖。
[Abstract]:Objective to study the expression and significance of proliferating cell nuclear antigen (PCNA) and p53 in atrophic parotid gland regeneration from gene and protein levels. Methods 102 Wistar rats were divided into experimental group and control group. The parotid duct of experimental group was ligated. After 7 days of ligation, the parotid duct was recanalized in group A (n = 14). The histomorphologic changes of parotid gland were observed by hematoxylin and eosin (HEH) staining. The expression of PCNA and p53 in parotid gland was detected by real-time fluorescence quantitative polymerase chain reaction and Western blot. Results on the 714th day after ligation of parotid duct, apoptosis of acinus and proliferation of ductal cells were observed in the two experimental groups. The relative expression of p53 in the same group was higher than that in PCNA. The relative expression of PCNA and p53 in group B reached the peak on the 5th day after the recanalization of the ductus, compared with the control group. The relative expression of PCNA and p53 decreased gradually in group A on the 21st day after recanalization, and close to the control group on the 28th day after recanalization. Conclusion the relative expression of p53 was increased after ligation of parotid duct, which induced apoptosis of parotid gland acinar, and PCNA expression increased gradually after catheter recanalization, decreased after peak value, and promoted the proliferation of acinus.
