星形膠質(zhì)細(xì)胞活化在心理應(yīng)激影響大鼠咬肌痛覺(jué)敏感中的作用

【摘要】:目的:探討心理應(yīng)激狀態(tài)下咬肌痛覺(jué)敏感增高現(xiàn)象與三叉神經(jīng)脊束核尾側(cè)亞核(Vc)內(nèi)星形膠質(zhì)細(xì)胞活化的關(guān)系。方法:健康雄性SD大鼠40只,隨機(jī)分為:空白對(duì)照組和應(yīng)激3、7、14 d組(n=10),實(shí)驗(yàn)組大鼠經(jīng)束縛應(yīng)激相應(yīng)時(shí)間后,采用曠場(chǎng)實(shí)驗(yàn)和高架十字迷宮實(shí)驗(yàn)觀察各組大鼠焦慮、抑郁樣情緒;Von Frey絲刺激檢測(cè)各組大鼠咬肌的機(jī)械性痛閾;免疫組化染色和Western-blot檢測(cè)Vc內(nèi)星形膠質(zhì)細(xì)胞活化標(biāo)志物神經(jīng)膠質(zhì)酸性蛋白(GFAP)表達(dá)的變化。結(jié)果:①束縛應(yīng)激7、14 d組大鼠在曠場(chǎng)中央停留時(shí)間和中央活動(dòng)總路程,以及在高架十字迷宮開(kāi)放臂中的滯留時(shí)間百分比和進(jìn)入開(kāi)放臂次數(shù)百分比均較空白對(duì)照組明顯降低(P0.05);②束縛應(yīng)激7、14 d組大鼠咬肌機(jī)械性痛閾均較空白對(duì)照組明顯降低(P0.05);③束縛應(yīng)激7、14 d組大鼠Vc內(nèi)GFAP的表達(dá)陽(yáng)性率及其蛋白的表達(dá)水平均明顯高于空白對(duì)照組(P0.05)。結(jié)論:慢性束縛造成的心理應(yīng)激能導(dǎo)致Vc內(nèi)星形膠質(zhì)細(xì)胞的活化,可能參與了心理應(yīng)激導(dǎo)致的咬肌痛覺(jué)敏感性的增高。
[Abstract]:Aim: to investigate the relationship between the increased pain sensitivity of masseter muscle and the activation of astrocytes in the caudal subnucleus of trigeminal nucleus under psychological stress. Methods: forty healthy male Sprague-Dawley rats were randomly divided into two groups: blank control group and control group. The rats in the experimental group were subjected to restraint stress for the corresponding time. The anxiety of the rats in each group was observed by open field test and elevated cross maze test. The mechanical pain threshold of masseter muscle was detected by Von Frey filament stimulation, and the expression of glial acidic protein (glial acidic protein) in VC was detected by immunohistochemistry and Western-blot. Results the rats in the 7d restraint stress group stayed in the center of open field and the total distance of central activity. The percentage of retention time and the number of times entering the open arm in the open arm of the elevated cross maze were significantly lower than those in the blank control group. The mechanical pain threshold of masseter muscle in the 714-day restraint stress group was significantly lower than that in the blank control group. The positive rate of GFAP expression and the expression level of GFAP protein in VC in the control group were significantly higher than those in the control group (P 0.05). Conclusion: psychological stress induced by chronic restraint may lead to the activation of astrocytes in VC and may be involved in the increase of pain sensitivity of masseter muscle induced by psychological stress.
