【摘要】：目的：觀察非綜合征型唇腭裂易感基因TGFβ3、IRF6、MSX1在小鼠胚胎唇和腭形成時(shí)期的表達(dá)情況，探索其在腭裂畸形中的作用機(jī)制，為后續(xù)實(shí)驗(yàn)及臨床實(shí)驗(yàn)提供實(shí)驗(yàn)依據(jù)。 方法：1.建立維甲酸誘導(dǎo)小鼠腭裂模型，在C57BL/6N孕鼠第10個(gè)妊娠日(Gestationday,GD),實(shí)驗(yàn)組按照100mg/kg體重對孕鼠進(jìn)行維甲酸一次性灌胃，對照組給予等體積植物油。分別于GD12天、GD13天、GD14天、GD15天，脫頸處死孕鼠，剖宮取胚鼠。 2.采用實(shí)時(shí)熒光定量PCR（quantitative real-time polymerase chain reaction, QRT-PCR）技術(shù)和免疫組化的技術(shù)方法檢測TGFβ3、IRF6、MSX1基因在胚胎GD12天、GD13天、GD14天、GD15天mRNA和蛋白的表達(dá)和分布情況。 結(jié)果：1.mRNA表達(dá)：TGFβ3mRNA的表達(dá)量，實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05）；實(shí)驗(yàn)組與對照組相比，實(shí)驗(yàn)組均高于對照組，其中GD14天差異有統(tǒng)計(jì)學(xué)意義（p0.05）。IRF6mRNA的表達(dá)量，實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05）；實(shí)驗(yàn)組與對照組相比，除GD15天實(shí)驗(yàn)組高于對照組外，其他各組均低于對照組，其中GD14天差異有統(tǒng)計(jì)學(xué)意義（p0.05）。MSX1mRNA的表達(dá)量，實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，除GD13天降低，其他各組均升高，差異均無統(tǒng)計(jì)學(xué)意義；實(shí)驗(yàn)組與對照組相比，除GD15天外，實(shí)驗(yàn)組均高于對照組，其中GD13天、GD14天差異有統(tǒng)計(jì)學(xué)意義(p0.05)。 2.蛋白表達(dá)：TGFβ3蛋白表達(dá)，，實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，其他各組均升高差異有統(tǒng)計(jì)學(xué)意義（p0.05）；實(shí)驗(yàn)組與對照組相比，實(shí)驗(yàn)組均高于對照組，除GD13天外其他各組均差異有統(tǒng)計(jì)學(xué)意義(p0.05)。IRF6蛋白表達(dá)：實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高，除GD14天其他各組差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，其他各組均升高，除GD15天其他各組差異有統(tǒng)計(jì)學(xué)意義（p0.05）。實(shí)驗(yàn)組與對照組相比，除GD15天實(shí)驗(yàn)組高于對照組外，其他各組均低于相應(yīng)對照組差異有統(tǒng)計(jì)學(xué)意義（p0.05）。MSX1蛋白表達(dá)，實(shí)驗(yàn)組中各組與GD12天相比，其他各組均升高，除GD15天其他各組差異有統(tǒng)計(jì)學(xué)意義（p0.05），對照組中各組與GD12天相比，除GD14天升高差異無統(tǒng)計(jì)學(xué)意義，其他兩組均降低差異有統(tǒng)計(jì)學(xué)意義（p0.05）；實(shí)驗(yàn)組與對照組相比，實(shí)驗(yàn)組均高于對照組，除GD12天其他個(gè)組差異有統(tǒng)計(jì)學(xué)意義（p0.05）。 結(jié)論：1.TGFβ3基因mRNA和蛋白表達(dá)水平的上調(diào)，可能與胚胎時(shí)期唇和腭的形成有關(guān)。 2.IRF6基因mRNA和蛋白在GD12～GD14天的低表達(dá)水平，異常調(diào)控腭突的生長發(fā)育，而導(dǎo)致唇腭裂的發(fā)生。 3.MSX1基因mRNA和蛋白的高表達(dá)水平，異常調(diào)控了腭突的生長發(fā)育，導(dǎo)致腭裂的發(fā)生。
[Abstract]:Objective : To investigate the expression of TGF - 尾3 , IRF6 , MSX1 in the development of mouse embryonic lips and palate by observing the expression of TGF - 尾3 , IRF6 and MSX1 in the development of mouse embryonic lips and palate , and to provide experimental basis for the follow - up experiment and clinical experiment .

Methods : 1 . To establish a model of mice with cleft palate induced by retinoic acid . The rats were given equal volume of vegetable oil on the 10th day of gestation ( Gestationday , GD ) in the experimental group . The control group was given equal volume of vegetable oil . GD12 , GD13 , GD14 , GD15 , pregnant rats were sacrificed .

2 . The expression and distribution of mRNA and protein in GD12 , GD13 , GD14 and GD15 were detected by quantitative real - time polymerase chain reaction ( QRT - PCR ) and immunohistochemistry .

Results : 1 . mRNA expression : TGF - 尾3 mRNA expression was significantly higher in the experimental group than that in GD12 ( p < 0.05 ) .
Compared with the control group , the experimental group was higher than that of the control group , among which GD14 had statistical significance ( p 0.05 ) . Compared with GD12 in the experimental group , the difference was statistically significant ( p < 0.05 ) .
In the experimental group , compared with the control group , all the other groups were lower than the control group except that the GD15 day group was higher than that of the control group , among which GD14 days had significant difference ( p < 0.05 ) . Compared with GD12 in the experimental group , the difference was statistically significant ( p < 0.05 ) .
Compared with the control group , the experimental group was higher than that of the control group , among which GD13 and GD14 had significant difference ( p < 0.05 ) .

2 . Protein expression : TGF - 尾3 protein expression , in the experimental group , the difference was statistically significant ( p < 0.05 ) compared with GD12 ( p < 0.05 ) , and in the control group , the difference was statistically significant ( p < 0.05 ) .
Compared with the control group , there was significant difference between the experimental group and the control group ( p < 0.05 ) . Compared with the control group , the other groups were higher than those in the control group ( p < 0.05 ) .
Compared with the control group , the experimental group was higher than that of the control group , and the difference between the experimental group and the control group was significantly higher than that in the control group ( p < 0.05 ) .

Conclusion : 1 . The up - regulation of TGF - 尾3 mRNA and protein expression may be related to the formation of lip and palate .

2 . The low expression level of IRF6 gene mRNA and protein in GD12 - GD14 could regulate the growth and development of palatine process , which could lead to cleft lip and palate .

3 . The high expression level of mRNA and protein of MSX1 gene regulates the growth and development of palate and leads to cleft palate .
