【摘要】:目的:觀察7-脫氫膽固醇還原酶(Dhcr7)基因在小鼠胚胎腭突發(fā)育中表達(dá)量的變化。方法:在體視顯微鏡下取E13.5、E14.5和E15.5的小鼠胚胎的腭突,免疫組織化學(xué)(IHC)觀察Dhcr7蛋白在腭突中的表達(dá)定位,逆轉(zhuǎn)錄式聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)法檢測Dhcr7 mRNA的表達(dá)變化,Western blot法檢測蛋白的表達(dá)變化。結(jié)果:Dhcr7主要表達(dá)在胚胎腭突間充質(zhì)細(xì)胞(EPM);E13.5、E14.5和E15.5 mRNA相對表達(dá)量(Dhcr7/β-actin)分別是0.692±0.084、0.595±0.051和0.369±0.061;E13.5與E14.5比較,P0.05;E13.5、E14.5分別與E15.5比較,P0.01;蛋白相對表達(dá)量(Dhcr7/GAPDH)分別是0.857±0.008、0.840±0.005和0.300±0.007,E13.5與E14.5相比,P0.05;E15.5分別與E13.5和E14.5相比P0.01。結(jié)論:Dhcr7在腭突發(fā)育中的關(guān)鍵階段有表達(dá),并且表達(dá)量與其生長發(fā)育有密切關(guān)系。
[Abstract]:Aim: to observe the expression of 7-dehydrocholesterol reductase (Dhcr7) gene in the development of mouse palatal process. Methods: the palatine process of mouse embryos of E13.5E14.5 and E15.5 were taken under stereoscopic microscope, and the expression of Dhcr7 protein in palatine process was observed by immunohistochemistry.
