雷尼酸鍶、強(qiáng)骨膠囊對骨質(zhì)疏松大鼠正畸牙移動藥物作用的研究

【摘要】:目的通過建立大鼠骨質(zhì)疏松、正畸牙移動實(shí)驗動物模型,同時給予雷尼酸鍶和強(qiáng)骨膠囊藥物干預(yù),檢測正畸加力后大鼠不同階段的脛骨近端骨密度、上頜第一磨牙移動距離以及牙周組織中胰島素樣生長因子1(IGF-1)和轉(zhuǎn)化生長因子1(TGF-β1)的表達(dá)情況。探討藥物干預(yù)后對骨質(zhì)疏松模型大鼠正畸牙移動的影響以及不同藥物的治療效果。為臨床骨質(zhì)疏松患者正畸治療選擇干預(yù)藥物提供實(shí)驗依據(jù)。方法實(shí)驗選取80只4周齡SD雄性大鼠,體重為(210±10)g,置于SPF級環(huán)境中飼養(yǎng),大鼠自由飲食水。80只大鼠隨機(jī)分成實(shí)驗對照組(A組),模型對照組(B組),雷尼酸鍶組(C組),強(qiáng)骨膠囊組(D組),每組各20只。實(shí)驗性喂養(yǎng)一周后B、C、D組建立骨質(zhì)疏松模型,給予維甲酸80mg/(kg·d)連續(xù)灌胃2周,A組給予等量蒸餾水作為對比。兩周后每組隨機(jī)處死5只大鼠行骨密度檢測(bone mass density,BMD),確定建模成功。建模成功后,給予D組強(qiáng)骨膠囊溶劑90mg/(kg·d);給予C組雷尼酸鍶溶劑400mg/(kg·d),進(jìn)行灌胃給藥;A組、B組則給予等量蒸餾水進(jìn)行對比。在用藥物干預(yù)治療兩周以后,所有大鼠安裝正畸裝置,加力后第7d、14d、21 d分批處死,每次隨機(jī)處死5只。取大鼠上頜第一磨牙及附近牙槽骨,測量加力側(cè)大鼠第一磨牙牙移動距離并進(jìn)行HE染色和免疫組化染色,應(yīng)用半定量法分析其牙周組織中IGF-1和TGF-β1的表達(dá)情況。結(jié)果1 B、C、D組大鼠脛骨BMD值均低于A組大鼠,差異有統(tǒng)計學(xué)意義(P0.05)。2同一加力時間段內(nèi)B組大鼠的第一磨牙移動距離高于D、C、A組大鼠,差異有統(tǒng)計學(xué)意義(P0.05)。3 C組和D組大鼠牙移動速度較B組有所減慢,差異有統(tǒng)計學(xué)意義(P0.05)。4免疫組織化學(xué)的檢測結(jié)果顯示,同一時間點(diǎn)C組和D組IGF-1表達(dá)較B組顯著增加,且7d和14d時表達(dá)較顯著,21天時表達(dá)有所降低,差異有統(tǒng)計學(xué)意義(P0.05);同一時間點(diǎn)C組IGF-1表達(dá)顯著強(qiáng)于D組,差異有統(tǒng)計學(xué)意義(P0.05)。5免疫組織化學(xué)的檢測結(jié)果還顯示,同一時間點(diǎn)C組和D組TGF-β1表達(dá)較B組顯著增加,且7d和14d時表達(dá)較顯著,21天時表達(dá)有所降低,差異有統(tǒng)計學(xué)意義(P0.05);同一時間點(diǎn)C組TGF-β1表達(dá)顯著強(qiáng)于D組,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論1實(shí)驗證實(shí)SD雄性大鼠給予維甲酸80mg/(kg·d)連續(xù)灌胃2周,可造成骨質(zhì)疏松模型。2骨質(zhì)疏松會使正畸大鼠的牙移動速度加快。3雷尼酸鍶與強(qiáng)骨膠囊均能降低骨質(zhì)疏松并使牙周組織中IGF-1和TGF-β1高表達(dá),提示成骨作用增強(qiáng)。4雷尼酸鍶對骨質(zhì)疏松藥物作用強(qiáng)于強(qiáng)骨膠囊。
[Abstract]:Objective to establish an experimental model of osteoporosis and orthodontic tooth movement in rats, and to measure the proximal tibia bone density of rats at different stages after orthodontic treatment with strontium ranidate and Qianggu capsule. The movement distance of maxillary first molar and the expression of insulin-like growth factor (IGF-1) and transforming growth factor (TGF- 尾 _ 1) in periodontal tissues. To investigate the effect of drug intervention on orthodontic tooth movement in osteoporosis model rats and the therapeutic effects of different drugs. To provide experimental evidence for the choice of intervention drugs for orthodontic treatment in patients with osteoporosis. Methods Eighty 4-week-old SD male rats, weighing 210 鹵10g, were fed in SPF environment. The rats were randomly divided into three groups: experimental control group (n = 20), model control group (n = 20), strontium raniate group (n = 20), group C (n = 20) and group D (n = 20). Osteoporosis model was established in group D after experimental feeding for one week. The rats in group A were given the same amount of distilled water for 2 weeks after continuous administration of retinoic acid (80mg/(kg d). Two weeks later, 5 rats in each group were randomly killed to measure bone mass density. After modeling successfully, group D was given 90mg/(kg dai, group C was treated with strontium ranionate, and group A was given the same amount of distilled water. After two weeks of drug intervention, all rats were installed with orthodontic devices, and were killed in batches on day 7, 14 and day 21 respectively, and 5 rats were randomly killed each time. The maxillary first molar and its adjacent alveolar bone were taken to measure the distance of tooth movement, HE staining and immunohistochemical staining. The expression of IGF-1 and TGF- 尾 1 in periodontal tissue was analyzed by semi-quantitative method. Results (1) the tibia BMD values of rats in group B were lower than those in group A, and the difference was statistically significant. The distance of first molar movement in group B was higher than that in group A during the same period of exertion. The rate of tooth movement in group C and group D was slower than that in group B, and the expression of IGF-1 in group C and group D was significantly higher than that in group B at the same time point, and the difference was statistically significant (P 0.054.The results of immunohistochemistry showed that the expression of IGF-1 in group C and group D was significantly higher than that in group B at the same time point). The expression of IGF-1 in group C was significantly higher than that in group D at day 7 and day 14, and the expression of IGF-1 in group C was significantly higher than that in group D at the same time point, and the results of immunohistochemistry showed that the expression of IGF-1 in group C was significantly higher than that in group D at the same time point. At the same time point, the expression of TGF- 尾 1 in group C and group D was significantly higher than that in group B, and the expression of TGF- 尾 1 in group C was significantly lower than that in group B at day 7 and day 14, the difference was statistically significant (P 0.05), and the expression of TGF- 尾 1 in group C was significantly higher than that in group D at the same time point (P 0.05). Conclusion 1Sprague-Dawley male rats were given retinoic acid (80mg/(kg d) for 2 weeks. 2 osteoporosis could accelerate tooth movement in orthodontic rats. 3. 3 strontium raniate and Qianggu capsule could decrease osteoporosis and increase the expression of IGF-1 and TGF- 尾 1 in periodontal tissue. The results suggest that the osteogenic effect is stronger than that of Qianggu capsule.
