【摘要】：目的研究碳酸飲料對(duì)乳恒牙更替期牙齒健康的影響作用及機(jī)制。方法 40顆犬牙釉質(zhì)樣本分別采用15μL可口可樂(可口可樂組)、雪碧(雪碧組)、純蘇打水(蘇打水組)和生理鹽水(生理鹽水組)浸泡,各10顆。測定處理后飲料中鈣、磷溶出濃度;分離牙乳頭干細(xì)胞,以添加可口可樂、雪碧、蘇打水和生理鹽水的條件培養(yǎng)基進(jìn)行細(xì)胞培養(yǎng),采用PCR法和Western blot法檢測核因子-κB受體活化劑配體(RANKL)、骨保護(hù)素(OPG)和維生素D受體(VDR)的mRNA和蛋白表達(dá)水平。結(jié)果各組鈣、磷溶出濃度(mmol/L)由高到低均依次為:雪碧組(3.679±0.114、0.089±0.008)、可口可樂組(2.217±0.109、0.036±0.005)、生理鹽水組(0.021±0.004、0.009±0.001)和蘇打水組(0.007±0.001、0.002±0.000)。各組RANKL的mRNA和蛋白表達(dá)差異均無統(tǒng)計(jì)學(xué)意義。蘇打水組和生理鹽水組OPG的mRNA和蛋白表達(dá)水平均高于可口可樂組和雪碧組(均P0.05),雪碧組與可口可樂組、生理鹽水組與蘇打水組間差異均無統(tǒng)計(jì)學(xué)意義。生理鹽水組VDR的mRNA和蛋白表達(dá)水平低于可口可樂組和雪碧組,高于蘇打水組(均P0.05)。結(jié)論碳酸飲料可能通過調(diào)控成骨相關(guān)基因表達(dá)及維生素D受體家族共同影響乳恒牙更替期的牙齒健康。
[Abstract]:Objective to study the effect and mechanism of carbonated beverage on the health of primary permanent teeth. Methods 40 canine enamel samples were soaked with 15 渭 L Coca cola (Coca Cola group), Sprite group (Sprite group), pure soda water (soda water group) and normal saline (normal saline group), 10 in each group. The calcium and phosphorus dissolving concentration in the treated beverage was determined, the dental papillary stem cells were isolated, and the cells were cultured on the conditioned medium supplemented with Coca-Cola, Sprite, soda water and normal saline. The expression levels of mRNA and protein of nuclear factor- 魏 B receptor activator ligand RANKL, osteoprotegerin (OPG) and vitamin D receptor (VDR) were detected by PCR and Western blot methods. Results the concentrations of calcium and phosphorus in each group ranged from high to low: Sprite group (3.679 鹵0.114), Coca Cola group (2.217 鹵0.109 鹵0.005), saline group (0.021 鹵0.004) and soda water group (0.007 鹵0.001 鹵0.002 鹵0.000). There was no significant difference in the expression of mRNA and protein in all groups of RANKL. The expression of mRNA and protein in OPG of soda water group and saline group were higher than that of Coca-Cola group and Sprite group (P0.05). There was no significant difference between Sprite group and Coca Cola group and normal saline group and soda water group. The expression of mRNA and protein in VDR of saline group was lower than that of Coca Cola group and Sprite group, and higher than that of soda water group (all P 0.05). Conclusion carbonated beverage may affect the dental health of primary permanent teeth by regulating the expression of osteoblast related genes and vitamin D receptor family.
