【摘要】：目的：研究經(jīng)不同途徑化療對兔舌VX-2移植性鱗癌區(qū)域轉(zhuǎn)移淋巴結(jié)癌細(xì)胞凋亡的影響。 方法：24只新西蘭兔舌緣種植VX-2腫瘤，隨機(jī)分為6組，每組4只。頸淋巴結(jié)最大橫徑大于5mm時(shí)開始化療。A組：經(jīng)舌動(dòng)脈灌注紫杉醇脂質(zhì)體和卡鉑注射液；B組：經(jīng)舌動(dòng)脈灌注游離紫杉醇和卡鉑注射液；C組：經(jīng)舌動(dòng)脈灌注5%葡萄糖溶液；D組：經(jīng)耳緣靜脈灌注紫杉醇脂質(zhì)體和卡鉑注射液；E組：經(jīng)耳緣靜脈灌注游離紫杉醇和卡鉑注射液；F組：經(jīng)耳緣靜脈灌注5%葡萄糖溶液。治療組給藥劑量紫杉醇7.5mg/kg、卡鉑13.0mg/kg�；熀�7天切除頸淋巴結(jié)進(jìn)行檢測：①透射電鏡下觀察轉(zhuǎn)移灶凋亡細(xì)胞微結(jié)構(gòu)的變化；②流式細(xì)胞儀檢測淋巴結(jié)轉(zhuǎn)移灶癌細(xì)胞凋亡率，ANOVA-LSD多重比較分析組間差異；③SP免疫組化檢測P53蛋白表達(dá)情況，計(jì)數(shù)資料采用χ2檢驗(yàn)和Fisher’s確切概率法。 結(jié)果：①各治療組均可見明顯的轉(zhuǎn)移癌細(xì)胞凋亡。②動(dòng)脈給藥組癌細(xì)胞凋亡率高于靜脈給藥組；紫杉醇脂質(zhì)體聯(lián)合卡鉑給藥組凋亡率高于游離紫杉醇給藥組。A組淋巴結(jié)縮小，，其余各組無縮小。各組癌細(xì)胞凋亡率分別為：A組（49.49±4.96）%、B組（29.65±3.61）%、C組（8.59±0.28%）、D組（21.05±1.65）%、E組（12.98±0.75）%、F組（8.47±0.30）%，除C組和F組外其余各組組間相比均有顯著差異（P＜0.05）。③P53蛋白在陽性率A組20.00%（2/12）、B組33.33%（4/12）、D組41.67%（5/12）、E組50%（6/12）、C組和F組75%（9/12），A組與C組和F組相比有顯著差異（P＜0.05），其他各組兩兩比較無統(tǒng)計(jì)學(xué)差異。 結(jié)論：靶動(dòng)脈灌注較全身靜脈灌注能更好誘導(dǎo)轉(zhuǎn)移淋巴結(jié)癌細(xì)胞凋亡、抑制腫瘤細(xì)胞增殖。紫杉醇脂質(zhì)體聯(lián)合卡鉑靶動(dòng)脈灌注能有效誘導(dǎo)區(qū)域淋巴結(jié)轉(zhuǎn)移癌細(xì)胞凋亡、抑制腫瘤細(xì)胞增殖。
[Abstract]:Aim: to study the effect of different ways of chemotherapy on apoptosis of metastatic lymph node cancer cells in transplanted squamous cell carcinoma (VX-2) of rabbit tongue. Methods Twenty four New Zealand rabbits with VX-2 tumor were randomly divided into 6 groups with 4 in each group. Group A: translingual infusion of paclitaxel liposome and carboplatin injection group B: translingual artery infusion of free paclitaxel and carboplatin injection group C: translingual artery infusion of 5% glucose Group D: paclitaxel liposome and carboplatin injection were perfused via auricular vein: group E: free paclitaxel and carboplatin injection were perfused via auricular vein: 5% glucose solution was perfused via auricular vein. The treatment group was given paclitaxel 7.5 mg / kg, carboplatin 13.0 mg / kg. Seven days after chemotherapy, cervical lymph nodes were resected to detect the changes of apoptotic microstructures of metastatic tumor cells under transmission electron microscope (TEM). 2 flow cytometry was used to detect the apoptosis rate of metastatic lymph node cancer cells and the difference of ANOVA-LSD multiplex analysis group was analyzed. The expression of p53 protein was detected by 3SP immunohistochemistry. The count data were analyzed by 蠂 2 test and Fisher's exact probability method. Results the apoptosis rate of metastatic cancer cells was significantly higher in arterial administration group than in intravenous administration group, and the apoptosis rate in paclitaxel liposome combined with carboplatin group was higher than that in free paclitaxel group.
