應(yīng)用QPCR驗(yàn)證口腔黏膜下纖維性變癌變分子標(biāo)記物的研究

【摘要】:目的:探求口腔黏膜下纖維性變(oral submucous fibrosis,OSF)癌變的特異性分子標(biāo)記物,為闡明口腔黏膜下纖維性變癌變關(guān)鍵機(jī)制及早期診斷提供理論基礎(chǔ)。方法:基于前期基因芯片的分析數(shù)據(jù),篩選出10個候選基因CXCL8、CXCL1、PTPRZ1、WDR66、DKK1、COX6A2、ATP1A2、TPM1、ABCA8、MIB1。采用實(shí)時熒光定量核酸擴(kuò)增檢測系統(tǒng)(QPCR)檢測在OSF癌變組織12例、非OSF癌變組織10例和正常組織8例中候選基因mRNA的表達(dá)。用2~(-ΔΔCT)法分析比較基因的相對表達(dá)量。結(jié)果:CXCL1、PTPRZ1在OSF癌變的頰粘膜組織中表達(dá)明顯高于非OSF癌變鱗癌組和正常對照組(P0.05);TMP1在OSF癌變的頰粘膜組織中表達(dá)明顯低于非OSF癌變鱗癌組和正常對照組(P0.05)。結(jié)論:CXCL1、PTPRZ1、TPM1基因?qū)SF癌變的早期診斷有意義,可望成為OSF癌變的早期診斷的分子標(biāo)記物。
[Abstract]:Objective: to explore the specific molecular markers of oral submucous fibrosis (submucous) carcinogenesis and to provide a theoretical basis for elucidating the key mechanism and early diagnosis of oral submucous fibrosis carcinogenesis. Methods: ten candidate genes (CXCL8, CXCL1, PTPRZ1, WDR66, DKK1, COX6A2AT1A2TPM1, ABCA8, MIB1) were selected based on the data of gene microarray analysis. The expression of candidate gene mRNA was detected in 12 cases of OSF cancerous tissue, 10 cases of non- cancerous tissue and 8 cases of normal tissue by real-time fluorescence quantitative nucleic acid amplification system. The relative expression of the gene was analyzed and compared by the method of 2 ~ (-)-螖 螖 CT. Results the expression of OSF in buccal mucosa with OSF carcinogenesis was significantly higher than that in non-OSF carcinomatous squamous cell carcinoma and normal control group. The expression of TMP1 in OSF carcinomatous buccal mucosa was significantly lower than that in non-OSF carcinomatous buccal mucosa and normal control group (P 0.05). Conclusion the OSF TPM1 gene may be useful in the early diagnosis of OSF carcinogenesis, and it is expected to be a molecular marker for the early diagnosis of OSF carcinogenesis.
